[Purpose] To study the protective effect and mechanism of compound angelica injection to the focal cerebral ischemia reperfusion.[Method] (1) the model of focal cerebral ischemia reperfusion in SD rats was established with suture-occluded method invented by Zea Longa.72 SD rats were randomly divided into six groups (sham operated group, focal cerebral ischemia reperfusion model group, three kinds of dose compound angelica injection groups, salvia miltiorrhiza injection group). Each group was 12 SD rats.in All the groups, scores of neurological deficit ,infarct volume, tumor necrosis factor-α(TNF-α)of blood serum , endothelin (ET)of blood plasm were observed at 24 hours reperfusion after 2 hours ischemia.(2) in All the groups , histopathological changes with HE staining under a microscopy, myeloperoxidase (MPO) and nitrogen monoxidum(NO) of the brain homogenate were obsearved at 24 hours reperfusion after 2 hours ischemia.[Results] (l)Compared with sham operated group, scores of neurological deficit and infarct volume of focal cerebral ischemia reperfusion model group were higher significantly (P<0.01). Compared with model group, scores of neurological deficit and infarct volume were lighted significantly in groups treated by compound angelica injection and salvia miltiorrhiza injection at 24 hours reperfusion after 2 hours ischemia (PO.01) .(2)Compared with sham operated group, tumor necrosis factor-a(TNF—a)of serum , endothelin (ET)of plasm, myeloperoxidase (MPO) and nitrogen monoxidum (NO) of the brain homogenate of focal cerebral ischemia reperfusion model group were increased (PO.01) .Compared with model group, all the treated groups decreased the biochemical indicators significantly (PO.01)[Conclusion] compound angelica injection could protect brain damage after focal crerbral ischemia reperfusion, and the mechanism of protection were seem to inhibit the expression of the cell factors about inflammation in the focal cerebral ischemia reperfusion.
