ObjectiveCerebral Vascular Disease (CVD) had the high attack rate, high disabling rate, high fatal character, which was the common disease and frequently encountered disease for the mankind, especially for the mid-aged people. With the aging population of the globe more and more increasing, the harm of CVD is increasingly outstanding. The most common was ischemic CVD. So the medical public pay more important attention to ischemic CVD. Ischemic CVD emphasized the early treatment, which took the present of thrombolysis therapy. But the thrombolysis therapy had a strict period window. If this period passed, it could induce the injury of ischemia-reperfusion. Astrocytes have intricate functions during the ischemia-reperfusion. Glial fibrillary acidic protein (GFAP) is the astrocyte maker protein. The expression of GFAP meant that astrocytes were activated. Used the MCAO model, this research investigates the effect of GFAP protein and the content of NO in the peripheral blood after CI to research the mechanism of early acupuncture treating resisting cerebral ischemical reperfusion injury after CI. MethodsSD rats were randomly divided into sham operation group, model(ischemia 0.5h reperfusion 24 hours), model(ischemia 0.5h reperfusion 48 hours), electroacupuncture treatment(ischemia 0.5h reperfusion 24 hours, treating 2 times), electroacupuncture treatment (ischemia 0. 5h reperfusion 48 hours, treating 3 times) and the normal control group. Used the suture in MCA to establish MCAO ischemia-reperfusing model. The remarks of nerve function deficit referring to Lundy remarking standards. The acupoints of BAIHUI and SHUIGOU were selected, stimulated with slow-fast wave for 30 minutes. The rats (eight rats from every group) were reperfused with 4 % paraformaldehyde after being anesthetized by 10% chloral hydrate, then taken out the brain. The brain tissue 2mm away from the chiasma was cut frontally, the middle part left, immersed in hard paraffin. The blood of other rats (six rats from every group) was col lected by extirpating eyealls, Which was put in the room temperation till thrombokinesis, centrifuging 10 minutes, separating serum, conservated in - 20 °C . Used immunohistochemistry SABC(spreptavidin-biotin-peroxidase complex) method to investigate the expression of GFAP and NO nitric acid reducing enzyme method in the ischemia brain after ischemia-reperfusion andelectroacupuncture. In the 40 times objects glass, astrocytes of GFAP positive reaction were analysis of accounts. The data of NO reaction was collected. All the datum were processed by SPSS11. 0 statistics software package and all results were demonstrated by mean±standard deviation. Results1. MCAO and reperfusion injury can distinctly do harm to rats' activity; electroacupuncture can prominently improve its activity after ischemia.2. The expression of GFAP in the rats' ischemia brain increases after ischemia-reperfusion. The content of NO in the serum increases after ischemia-reperfusion.3. Then electroacupuncture can make the expression of GFAP, NO in the rats' ischemia brain decrease.Conclusion1. Reactiving the AST to injury the brain issue after the cerebral ischemia-reperfusion.2. Electroacupuncture can make the expression of GFAP, NO in rats' ischemia brain the very level, then conduce to the protective actions to the brain.3. The mechanism of early acupuncture treating can effectively intervene the injury of CI, is through the immunal regulation to the inflammation reaction after cerebral ischemia.
