Adverse Effects And Its Mechanism Of Environmental Endocrine Disrupting Chemicals P-nonylphenol(p-NP) On Sertoli Cells In Testis Of Sprague-Dawley Rats

Alkylphenols are applied widely as the rubber additives surfactants in domestic and industrial products, antioxidants and so on. As it's decomposer, nonylphenol(NP) has a steady chemical characteristics and exist widely in environment. As it's molecule structures are similar as 17beta-estradiol in human body, and it can combine with estradiol receptor simulating it, disrupt endocrine system and induce health damage effect. Many studies on environmental endocrine disrupting chemicals approved that NP have weaken estradiol functions. Since 90s of the previous century, the studies on the health damage effect induced by environmental endocrine disrupting chemicals have become a hotspot in the field of biomedicine, more and more reports about the health damage effect induced by environmental endocrine disrupting chemicals have come forth. But there are few reports on the mechanism of NP inducing the adverse effect on the reproductive function of F1 male SD rat .In this study we chose p-NP as object, explore the effect of NP on the function of testis Sertoli cells of male SD rat and it's mechanism by tests in vivo and in vitro.The female rats were exposed to nonylphenol from P7 to lactation period ,and observe the effect of nonylphenol on the function of Sertolicells of F1 male SD rat after lactation period. It shows that nonylphenol can inhibit the expression of pan cadherin in testis of male SD rat ,at the same time it can inhibit the expression of Vimentin in testis Sertoli cells of male SD rat after lactation period, further more the expression of pan cadherin and Vimentin tends to decrease with the increase of exposure dose . In all treated with NP groups the expression of Vimentin decrease compared with witch of control group statistically(p<0.05) while In treated with 100 mg/kg and 200mg/kg NP groups the expression of pan cadherin decrease compared with witch of control group statistically(p<0.05).Stop giving nonylphenol to females after lactation period, and observe the effect of nonylphenol on the function of Sertoli cells of F1 male SD rat in sex mature period. It shows that NP can weaken the expression of both Vimentin and pan cadherin in Sertoli cells of F1 male SD rat in sex mature period. In 200mg/kg NP group the expression of both Vimentin and pan cadherin are different from witch of control group statistically(p<0.05); NP can also weaken the expression of IL-6 mRNA and ABP mRNA in Sertoli cells of F1 male SD rat in sex mature period. In 100mg/kg NP group and 200mg/kg NP group the the expression of IL-6 mRNA and ABP mRNA are different from with of control group statistically (p<0.05); And NP has no significant effect on the expression of STAT3 in Sertoli cells of F1 male SD rat in sex mature period.Isolated and cultured the rat Sertoli cells in vitro , design the direct exposure doses(10-5mol/L, 10-6mol/L, 10-7mol/L, 10-8mol/L NPgroup, impregnant control group and blank control group) by MTT . The result shows that NP can effect on the Sertoli cells produce androgen binding protein in isolated rat Sertoli cells. There is no statistic difference between control group and 10-7mol/L, 10-8mol/L NP groups in the expression of ABP mRNA .But there is significant differences between control group and 10-5mol/L , 10-6mol/L NP groups(p<0.05).In 10-5mol/L and 10-6mol/L dose group NP effects on the expression of EL-6 mRNA in rat Sertoli cells significantly(p<0.05) and in 10-5mol/L, 10-6mol/L, 10-7mol/L dose group it effects on the expression of Vimentin in rats Sertoli cells significantly(p<0.05). Under the exposure doses of this study NP can weaken the expression of STAT3 and pan cadherin in rats Sertoli cells statistically (p<0.05).The results of this study shows that following giving high dose NP to female rats by gavage it appears obvious reproductive and developmental toxicity in F1 male rats. The Sertoli cells of F1 male rat in both after lactation period and sex mature period are damaged, and the possible mechanism is that NP has adverse effect on the function of Sertoli cells and induces reproductive and developmental toxicity.
