Chromosomal number abnormality (chromosomal instability) is one of the characteristic changes in tumor cells. The mechanism can't be understood clearly though it has been found for a long time. Since sister chromosome separation mechanism was founded in the recent years that securin/PTTG is degradated by APC(anaphase-promating complex),after its degradation,separase/ESPl activates, activing ESP1 then hydrolyzes cohesin/SCC1, and sister chromosome separates in the centromere in the mitosis anaphase. There is no reports indicating the mechanism in sister chromosome separation whether is as the same in tumor cells as the normal cells.And there is no reports whether sister chromosome separation correlative genes have some relationships to the chromosomal number abnormality. This study tries to discuss the question about sister chromosome separation correlative genes' expression mode in the different phases of A549, SPC-A-1 tumor cells, and to clarify the relationship to the chromosomal number abnormality.Objective:To discuss the expression regularity of sister chromosome separation correlative genes in the tumor cell lines, explore their roles in the process of the chromosomal number abnormality .Methods:1. Acquire the different phases of the 3 kinds of cells through double thymine synchronization.2. Detect SCC1,ESP1,PTTG,Cyclin B1 expressions in the different phases of A549,SPC-A-1,MRC-5 cell lines by FQ- PCR.3. Detect SCC1,ESP1,PTTG,Cyclin B1 expressions in the different phases of A549, SPC-A-1,MRC-5 cell lines by ISH.Results:We observed that SCC1,ESP1,PTTG,Cyclin B1 express regularly in the near diploidand aneuploidy cell.The regularity is ESP1 and Cyclin B1 express in the G2/M phase higher er than S phase, SCC1 express in the G2/M phase lower than S phase in near diploid and aneuploidy cell lines.But PTTG expresses different between near diploid and aneuploidy cell lines.In the near diploid cell line,we can't detect the PTTG in the S,G2/M phases.On the contrary, securin expresses in the G2/M phase lower than S phase in aneuploidy cell lines.Conclusion:We observed that SCCl,ESP1,PTTG,Cyclin B1 express regularly in the near diploid and aneuploidy cell lines;PTTG which inhibits ESP1 activation expresses higher in the aneuploidy cell lines, this maybe has some relationships to chromosomal number abnormality.
