【Background】The hypoxia was an important enviroment stimulation factor , it could cause the organism synthesize a series of unique stress proteins to adapt misstatus. A special component had been found in blood of the human which entered to the hypoxia enviroment, there was a character absorption cusp with light spectrum ,spectrum information was different from the substance correlation with the mountain sickness which had been knew. There was correlation between the quality and dose of special component and the mountain sickness, γ=0.93.Identical structural substance would have same hydrogen spectrum expression. It was confirmated that the absorbance level of the special substance in Wistar rat and rabbit at drugged state was lower than that at wakefulness state. The β-endorphin which had important adjustment to nerve and immune system could inhibit the special substance to excrete. As it had similar function to the anti-hypoxia stress compound, we called it the hypoxia stress peptide(HSP). Then the blot detecting method was created, it was simple easy to detect the HSP for the grass-roots unit.The discover of HSP is an important achievement in aliplano medicine,but we are notdefinitude of the molecule compose and the relationship with mountain sickness. Along with the progress of the research if proteomics,many new study methods have widly been used in the research if the structure and function of pretein. mass spectrometry(MS) ,1H-nuclear magnetic resonance spectroscopy(1H-NMR) ,13C-nuclear magnetic resonance spectroscopy(13C-NMR) and fourier transform infrared spectrometry(FTIR),Ultravlolet spectrophotometry (UVS) have been paid more attention in the structure analyse. 【Objective】To purification the HSP from the sufferer of AMS and experiment animal,analysis the molecular formula, molecular structure , compose of amino acid with mass spectrometry(MS) ,1H-nuclear magnetic resonance spectroscopy(1H-NMR) ,13C-nuclear magnetic resonance spectroscopy(13C-NMR) and fourier transform infrared spectrometry(FTIR),Ultravlolet spectrophotometry (UVS). Research the relationship with mountain sickness by the way of inspect the level of HSP of some kinds of montain sickness man. 【Methods】1. To purification the HSP from 29 AMS sufferers and experiment rabbits through the way of high speed centrifugal and ion exchange chromatography. Measure the molecular weight of HSP with Finnigan Liquid chromatography-mass spectrometry(LC-MS). Measure 1H-nuclear magnetic resonance spectroscopy(1H-NMR) with Varian—INOVA400 NMR. Measure 13C-nuclear magnetic resonance spectroscopy(13C-NMR) with BRUKER AVANCE 600 NMR. Analyse the molecule structure of HSP combine the FITR and UVS. 2,To draw the serum from the health and patient of the rapidly in altiplano man ,high altitude pulmonary edema (HAPE ) sick, wounded sufferer and burthen exercise man. Them detect the HSP with the method being created by us. 【Results】1.Information of the spectrum:the definite radical was existed at the corresponding position from the FTIR diagram, the molecular weight was 1593.7 from the MS diagram. The molecular formula is C78O13N17H114, the possibility compose of amino acid is Phe ,Thr ,Arg ,Lys ,Trp ,Tyr, Pro,Ile, Val. 2.The content of the HSP were rapidly increaseed in the rapidly in altiplano man (P<0.05),them desend after adapt the altiplano , The content of the HSP were also greatly increaseed high altitude pulmonary edema (HAPE ) sick and wounded sufferer(P<0.05), The content of burthen exercise man is correlation with the burthen. 【Conclusion】The excessive subject is acrossed is this research, The HSP was exited in the fluid of the cowed of easy to sickness , The character of the HSP was differ from other substance, the structure information of the HSP had been provided by the mass spectrometry(MS),1H-nuclear magnetic resonance spectroscopy(1H-NMR),13C-nuclear magnetic resonance spectroscopy(13C-NMR) and fourier transform infrared spectrometry (FTIR),Ultravlolet spectrophotometry (UVS),. There was close correlation between the HSP and the mountain sickness by detected the HSP in the serum of the patients of the health and patient of the rapidly in altiplano man ,high altitude pulmonary edema (HAPE ) sick, wounded sufferer and burthen exercise man..
