Express Of TheTGF-α, TGF-β And IL-8mRNA On Tissues Of Small Intestine Of Rats With Obstructive Jaundice

objective: The barrier of the intestinal mucosa would be impaired severely because of the obstructive jaundice, and consequent enteropathica endotoxemia and the displacement of the Intestinal tract bacterium were the main reason of the complication and death of the patients with the obstructive jaundice who surround the operation. This experiment will study the pathophysiological mechanism of impairment of the barrier of intestinal mucosa through determinating the quantity of TGF- α~+, TGF-β,and IL-8mRNA~+ cells and the endotoxin of the portal vein. And ti would be provide the theory basis to the clinical prevention and cure. Methods: 48 mail Wistar Rats were divided into pseudo-operation group and obstructive jaundice group, and were determinated the quantity of direct reating bilirubin and endotoxin in portal vein at the 4th, 7th, 14th day after operation and were determinated the quantity of TGF- α~+, TGF-β~+ and IL-8mRNA~+ cells of the intestinal tissue. Resultant: (1) The quantity of direct reating bilirubin and endotoxin in obstructive group were much higher than the pseudo-operation group (P<0.01); the quantity of endotoxin in 4 days' obstructive jaundice group was lower than the 7 days' group and 14 days' group (P<0.01); and there was no significant difference in the obstructive jaundice group about the quantity of direct reating bilirubin (P>0.05) . (2) The quantity of TGF- α~+, TGF-β~+ and IL-8mRNA~+ cells in obstructive jaundice group were lower than the pseudo-operation group (P<0.01) . And in obstructive jaundice group the quantity of TGF- α~+, TGF-β~+ and IL-8mRNA~++ cells were lessoned with the length of the day (P<0.05). conclusion: (1) TGF-α of the lamina propria mucosae would lessoning while the endotoxin in portal vein was heightening, so, the impairment of barrier of the intestinal mucosa was caused .(2)The TGF- β~+ and IL-8mRNA~+ Inhibition of organic immunological function descent was occurred the early time of obstructive jaundice, and aggravated with course. So it is necessary to use the antibiotics during the time of operation surrounding. (3) Protection on the, intestinal mucosa, heightenthe organic immunity and lesson the concentration of the endotoxin in the intestinal tract during the time surrounded the operation were the important link about the heightening the organic immunity and the prevention the high-rate infection after the operation...
