Objective:To investigate the new way of treating tubulointerstitial fibrosis (RIF) by collateral disease doctrine and the mechanism of adjusting of extracellular matrix (ECM) by the Regulating Collateral and EliminatingTurbidity Liquid (REL). Methods: 60 wistar rats was divided into 4 groups randomly :the normal control group(group Ⅰ ),the normal rat perfusing with REL group(group Ⅱ ),the operation model group (groupⅢ)and the operation model perfusing with REL group(groupIV).The model of RIF was established by unilateral ureteral obstruction (UUO)method,and five weeks later the kidney tissue were be taken for measuring the expression of the Collagen Ⅲ,Ⅳ Fibronectin (FN) and Transforming Growth Factor- β1 (TGF- β1) by the means of immunohistochemistry. Result: Comparing with the control group, the Collagen Ⅲ,Ⅳ and FN express remarkably increased in RIF rat, the factor due to fibrosis- TGF- β1 also express intensely . After perfusing the REL , comparing with operation model group the Collagen Ⅲ,Ⅳ and FN express weakly, the expression of TGF-β1 decreased. The degrees of fibrosis was remarkably ameliorated. Conclusion: The kidney collateral stasis is the primary pathological basis of RIF, blood-nourishing, blood-promoting, collateral-regulating, turbidity-eliminating therapy is effective on it. The REL can remarkably ameliorate the deposition of ECM of RIF rat. One of the mechanism maybe by inhabiting the expression of TGF- β1 reducing the deposition of ECM, then affecting the progression of RIF.
