Objective: To evaluate the efficacy of intranasal fluticasone propionate tothe histopathological changes in nasal mucosa of guinea pigs following a series ofirradiation.Methods:A total of 48 healthy guinea pigs were divided into two groupsrandomly: the irradiation group (n=24) , the irradiation and remedy group (n=24).The total of 20 animals of each group were exposed nasally to Co-60 acceleratorradial at 30Gy, quintic a week for two weeks. All of guinea pigs were treated overone month period with fluticasone propionate aqueous nasal spray 100 ug qd, At1d,2w,4w,8w,12w after irradiation four guinea pigs per one group were sacrificedrandomly, at each time point. Their middle nasal turbinate mucosa was observedunder light microscope, scanning electron microscope and transmission electronmicroscope.Results:The degree of degeneration and necrosis in the nasal mucosa wasmore severe in the irradiation group than in the irradiation and remedy groupunder the observation with Light microscope at the first day and the second week.Inflammatory cell infiltration was prevalent at that time, most of them areneutrophilic granulocyte. The repair of the nasal mucosa was prevalent at theeighth week and the twelfth week. the hyperplasia of fibrous tissue was far morethan in the irradiation group. The observation under scanning electron microscopedetected that areas of ciliary loss, falling off and different directions of the ciliawas more severe in the irradiation group. Secretion that covered on the nasalmucosa was easy to see in the irradiation group. The observation undertransmission electron microscope showed that nuclear pyknosis of ciliated cellsstarted after irradiation. Cytoplasmic vacuolation and expansion of intercellularspaces continued until the twelfth week. The change of ultrastructure was easy tosee in the irradiation group.Conclusion Fluticasone propionate aqueous nasal spray was effective inthe protection of guinea pigs'nasal mucosa which was irradiated.
