Objective:This topic proposed by animal experiment in rats, observe its effect on the rat nasal mucosa ciliary movement and mucous membrane irritation.To study the feasibility of nasal drug delivery by Yuxingcao, to explore the clinical values of Yuxingcao, and to develop a broader prospect.Method:Using rat nasal mucosa ciliary movement model method, the selection of30healthy common male rats randomly divided into five groups:normal saline group (blank group),Yuxingcao (for3,5,7days) group,1%sodium deoxycholate group (positive group).Bilateral nasal drug delivery, each by micro syringe. Observing the ciliary movement duration under optical microscope, Evaluating the influence on ciliary movement. According to pathological biopsy examination, by comparing the rat nasal mucosa stimulation points, evaluation on drug stimulating effects of nasal mucosa on rats.Results:1.By observing the ciliary movement duration under optical microscope, the results show that the ciliary movement is active, the cilium no loss, and cilium row neatly in physiological saline group, Yuxingcao (for3,5,7days) group; mucosa of rats damaged severely, the surface clutter, cilia fell out, see bare base, visible around a lot of exfoliated cells in sodium deoxycholate group. The ciliary movement duration of sodium deoxycholate group is shorter than normal saline group, the difference was statistically significant (P<0.05), the ciliary movement duration of Yuxingcao (for3,5,7days) group is shorter than the normal saline group, but there was no statistically significant difference (P>0.05), and we can consider that Yuxingcao has no effect on ciliary movement.2. By observing the nasal mucosa tissue pathological slices under optical microscope, according to the scoring criteria of mucosa pathological changes and inflammatory cells invasion situation,the pathological observation by HE dyeing show that the epithelium of the physiological saline group is complete, without inflammatory cell infiltration, hemorrhage, necrosis and edema; epithelial of Yuxingcao (for3,5,7days) group some basic is complete,with a small amount of inflammatory cells infiltration, hemorrhage, necrosis, mild edema; epithelial of1%sodium deoxycholate group is incomplete, with more inflammatory cells infiltration, focal hemorrhages, local necrosis, moderate edema. The nasal mucosa stimulation points of sodium deoxycholate group is higher than the normal saline group, the difference was statistically significant (P< 0.05), the nasal mucosa stimulation of Yuxingcao (for3,5,7days) group is higher than that of normal saline group, but there was no statistically significant difference (P>0.05), so we can consider that Yuxingcao has no stimulation on nasal mucosa.Conclusion:Yuxingcao has no effect on ciliary movement,and no stimulation on nasal mucosa of rats.
