Nowadays the most antibiotics are rapidly absorbed,reaching the highest concentration in blood quickly and meeting the need of sterilization in a short time. But the defect of it is only persisting a little time, and it has to be taken regularly. This greatly brings inconvenience to human beings and agony to the patient. . Sustained preparation can release drug slowly in delivering, and last the concentration of the drug in blood in a definite range in a period of time. Poly vinyl alcohol (PVA) is a kind of sustained preparation. PVA fiber film (PVAFF) forms PVA hydrogel after absorbing water. Because it is a three-dimensional network structure, drug can be released through it slowly. Chitosan has the function of antibacterial,hemostasis and accelerating the wound healing beside its releasing slowly, it can alleviate infection, adhesion and scar formation in the wound. In this study, the PVA hydrogel (PVAH) was formed through PVAFF immersed in liquid with antibiotics and chitosan (10% concentration), in order to cure the infection of wound and vaginosis, and assess its effet. The purpose of this research was offering a kind of preparation for releasing slowly, retaining wetly, restraining inflammation, adhesion and scar formation in clinical infection. 1. Inhibiting test of fungal growth with Kujinpi compound The function of Kujinpi compound was determined for superficial and inner fungi through the size of fungal inhibitory ring. the result showed that inhibiting fungal growth of Kujinpi compound was effective, and the best concentration was 10%. 2. Inhibiting test of bacteria and fungi growth with PVAH in vitro PVAH was made of PVAFF with diluent cerfuroxime sodium or Kujinpi compound and 10% chitosan. Then the inhibitory test of bacterial and fungal growth was done with PVAH and a round sheet of paper. Staphylococcus aureus, Escherlchia coli and Candida albicans were selected. The result showed that the time of PVAH inhibiting bacterial and fungal growth was longer than that of round paper. 3. The study of antibiotic releasing concentration-time Antibiotic releasing concentration-time curve was drawn according to OD value under 520 nm wavelength after PVAH inhibiting Staphylococcus aureus growth in 24h. PVAH was placed in mouse back, and taken off in different time, then the inhibiting effect was tested. The releasing ratio of PVAH in different time was worked out according to OD value in the curve. The result showed that antibiotic in PVAH was released 75% in 30h, the most in 42h, and was released completely in 48h. 4. The effet assessing of curing the mouse skin infection with PVAH after setted up the model of skin infection, the mouse skin infection was cured with gauze in 0.9% NaCl (negative controll), gauze in cerfuroxime sodium (positive control) and PVAH in cerfuroxime sodium. Compared with positive control, the ratio of no bacteria was higher in PVAH group. After 7 days, there were no statistical difference (P>0.05) in the wound contraction and the healing of skin ulcer, but distinctly showed statistical difference (P< 0.01) after 14 days. 5. The effect of PVAH on vaginosis rats The bacterial and fungal vaginosis of rats was cured with PVAH by changing drugs per 2d after done model. The result indicated that the number of no bacterial rat was more in PVAH group, and inflammation was alleviated in pathological inspect distinctly. We can draw conclusion 1.Outer and inner fungi can be inhibited by compound Kujinpi. 2.In the antibacterial experiment in vitro, inhibitory time of bacteria and fungi growth by PVAH is longer than round paper, as can prove that PVAH has sustaining funtion. 3.The drug in PVAH was released 75% in 30h, 92% in 42h, and was released completely in 48h in the mice's skin. 4.The effet of PVAH is better than ordinary gauze distinctly in curling the mice's skin infection.
