Pharmacokinetics, an important branch of pharmacology, takes plasma concentration-time curve (AUC) as its focus of study. The drug's concentration in plasma can reflect its status in target organs and many research works are available for reference. Usually plasma is used as the sample in pharmacokinetics study. Because of the powerful separating ability of high performance liquid chromatography (HPLC), the high sensibility and specificity of mass spectrometry (MS), and the detailed information of target compounds' structures obtained by collision induced dissociation (CID) or MS/MS, HPLC/MS technique is playing a more and more important role in pharmacokinetics study. Nowadays, many drugs, which have good therapeutic efficacy with small dose, are of high polarity and difficult to volatilize. In addition, with the development of biotechnology, bioactive compounds of strong polarity, such as polypeptide and high-molecular-weight proteins, are widely used to treat diseases.
