Regulatory function is provided with T_r cells ,which are divided as CD4+CD25+Tr cells, Trl and Th3 and manifold subtype according to difFerrantiation between surface marker, producing cell factor, effective machanism. At present, one of studiedly-clear cells is CD4+CD25+Tr.There adsum 5-10% to express CD25 molecule in ordinary person and small rats peripherial blood and spleen . CD4+CD25+Tr cells play an immportant role on inducing self tolerance, remaining T cells self steady, avoiding self immunity diseases and contradicting translantition rejection. According clinical result, CD4+CD25+ in patients who developed aGVHD is high compared with that undeveloped. It is apparent that CD4+CD25+ in patients who developed aGVHD increasing may suppress proliferation of CD4+ or CD8+ cells . So, it is immportant to establish a approach toculture and proliferation CD4+CD25+ cells, which is clinically-feasable and practically-convenient.Objective: To study the relationship between CD4+CD25+ and aGVHD and the expansion of CD4+CD25+ population in vitro.Method: CD4+CD25+ population were detected by FACS in patients with or without aGVHD after Allo-HSCT on +21, +25, +29, +33, +37, +41, +45 day . Peripherial T cells were purified by nylon cotton column,and cultured in RPMI1640 with anti-CD3(5ug/mL and EL-2, 500U/mL), CD4+CD25+, CD28+ cell were detectd by FACS after 14 days of culture.FasL were detected by RT-PCR.Result: CD4+CD25+ in patients who developed aGVHD is higher compared with that undeveloped.
