Investigation On The Influence On AGVHD After Transplantation Of The Fludarabine Preserved Cells Of Donor Rats

AIM:To explore a new approach of preventing and treating of aGVHD by investigating the incidence and severity of aGVHD after allogeneic bone marrow transplantation (Allo-BMT) of the Fludarabine preserved cells of donor Rats.Methods: To establish a rat allogeneic transplantation model,donors were Wister rats,recipients were SD rats.Twenty receptor SD rats were assigned randomly to three groups of A,B and C.All of the three groups received total body irradiation(TBI) 6.0Gy as myeloablative pre-conditioning.Four hours later,rats of group A were injected saline as control group, Rats of group B were injected with donor bone marrow and spleen cells via tail vein ,while rats of group C were injected with donor bone marrow and spleen cells cultured with fludarabine for 24h in vitro via tail vein as test groups.Afterward we observed haematopoietic reconstitution and aGVHD of three groups of receptor rats. Results: After transplantation,groups of B and C rats regained their haematopoietic recovery on day (18.50±1.05) and (15.17±1.47) respectively,statistical difference between the two groups were significant(P<0.05). Group A rats did not regained their haematopoietic recovery until died.Clinical aGVHD scores of group B is(7.33±1.63) ,while group C is(4.67±1.23),statistical difference between the two groups were significant(P<0.05). According to pathological ranking of liver and colon,group B had IV-V aGVHD chiefly,and group C had II-III aGVHD chiefly, statistical difference between the two groups were significant(P<0.05).Conclusions: In the rat allogeneic haemopoietic stem cell transplantation model,fludarabine preserved stem cell of donor rats can relieve aGVHD of receptor rats significantly,it provides a new way of protecting from aGVHD.
