Objectives Muscle glycogen synthase (GYSl) is the limiting-rate enzyme of glucose nonoxidative metabolism in the skeletal muscle. Polymorphism of GYSl gene is associated with high risks of insulin resistance in type 2 diabetes, and it may be a genetic marker of type 2 diabetes in some population. Our research object is the Han nationality population in Tianjin for a long time. There are 234 type 2 diabetes patients and 121 healthy control subjects in our study. The aim of this study is to investigate the prevalence of the M416V polymorphism of GYSl extron 10 and the Xba I polymorphism of GYSl intron 14 by PCR-RFLP technique. In addition, our aim is to find out the correlation between the two polymorphisms and insulin resistance in type 2 diabetes in the Han nationality population in Tianjin of China.Methods1. To collect the blood samples of type 2 diabetes patients and healthy control subjects.2. To extract and to purify the genomic DNA from the collectedblood samples.3. Using PCR-RFLP technique, the M416V polymorphism site ofGYSl extron 10 and the Xba I polymorphism site of GYSl intron 14 were screened in 234 type 2 diabetes patients and 121 healthy controls.4. To sequence the polymorphic gene found by RFLP technique.5. To analyse the clinical data.Results To the M416V polymorphism site of GYSl extron 10, 2 (0.9%) with the homozygote mutation, 53 (22.6%) with the heterozygote mutation were found in 234 type 2 diabetes patients. The V allele frequency is 12.2% in diabetes group. 3 (2.5%) with the homozygote mutation, 25 (20.7%) with the heterozygote mutation were found in 121 healthy controls. The V allele frequency is 12.8% in control group. There is no significant differences between two groups in the V allele frequency (p>0.05). In both diabetes group and control group, HOMA-IR index is higher in M416V ( + ) genotype subjects than that in M416V (-) genotype subjects. But compared between this two groups, it isn't seen the significant differences in statistically. In diabetes group, we subsequently compared the clinical data between M416V ( + ) and M416V (-) genotype subjects, and found there is significant differences in BMK P2PC TG> FIB between this two genotypes (p<0.05). To the Xba I polymorphism site of GYSl intron 14, all genotypes are A1A1 in both diabetes group and control group. A2 allele was no found in all subjects.Conclusions In the Han nationality population in Tianjin of China, the M416V polymorphism of GYSl extron 10 is associated with some indexes correlated insulin resistance. So it may partly cause to insulin resistance in the skeletal muscle of type 2 diabetes. The Xba I polymorphism of GYSl intron 14 is not associated with insulin resistance of type 2 diabetes in the Han nationality population in Tianjin of China.
