Objective: To investigate the regulating effects of transforming growth factor-β1 and basic fibroblast growth factor on the proliferation and collagen synthesis of human nasal polyps fibroblasts(NPFs).Methods: NPFs were dissociated and cultured in vitro from the excised nasal polyp from patients with chronic sinusitis with nasal polyps. The source of the cultured cells was identified by immunohistochemistry. NPFs was treating with different concentration of TGF-β1(1, 10, 50, 100 ng/ml), b FGF(1, 10, 50, 100 ng/ml) or both TGF-β1(10 ng/ml) and b FGF(10 ng/ml) for 24 h,72h,120 h. The proliferation of nasal polyps fibroblast was measured by Cell Titer 96® AQueous One Solution Cell Proliferation Assay(a), and the collagen III in the conditioned media was determined by ELISA.Results:(1) The cells migrated out from the tissue after tissue adherent to the culture vessel for 3 days, and the bottom of the culture vessel was covered with a continuous layer of cells in about 12 days. Immunohistochemical positive appraised waveform protein expression, and the cell culture was stable.(2) TGF-β1 and b FGF with various concentrations(1, 10, 50, 100 ng/ml) could not affect the growth of NPFs in 24 h clearly(P>0.05). But TGF-β1 with various concentrations(1, 10, 50, 100 ng/ml) could inhibit the growth of NPFs became more obvious while the phase was extended to 72 h or 120 h.On the contrary, b FGF with various concentrations(1, 10, 50, 100 ng/ml) could promote the NPFs growth. Compared with the control, the differences had reached statistical significance(all P <0.05). The combined application of TGF-β1 and b FGF shows significance antagonistic effect on the growth of NPFs compared with each single(P <0.05).(3) The type III collagen in NPFs supernatants gradually decreased after affected by variousconcentrations of TGF-β1(1, 10, 50, 100 ng/ml) for 120 h. And the result reversed affected by b FGF. Compared with the control, the differences had reached statistical significance(all P <0.05). The combined application of TGF-β1 and b FGF shows antagonistic effect on the collagen synthesis with each single(P<0.05).Conclusion: TGF-β1 could inhibit the NPFs growth and type III collagen synthesis, while b FGF could produce opposite results. There may be antagonistic effect between TGF-β1 and b FGF on the proliferation of NPFs,and it may play a important role on the tissue remodeling of CRS.
