Objective: To study the mechanism of breast carcinogenesis , we used peripheral blood of patients with breast cancer, through the measurement of the rate of anueploidy , S-phase fraction(SPF), Apoptosis(Apo), cyclin D1 and pl6 protein expression, and the influence of operation to these above markers.Methods: Using flow cytometry with propidium iodide staining and indirect immunofluorescence , cell proliferation indexes [the positive rate of anueploidy,S-phase fraction(SPF), Apoptosis(Apo)] and the expression of pl6 and cyclin Dl protein , thelymphocyte extracted from peripheral blood between 60 cases of breast cancer (beforeoperation and after operation in a week) and 20 cases of normal ones , were closely studied.Results: It was showed that the rate of anueploidy of breast cancer(43.33%) and DI(1.63±0.86) were obviously higher than those of normal ones(0.00%, 1.00±0.01) (p<0.01, p<0.05). In SPF, there was no significant difference between patients with breast cancer before operation (0. 34 ± 0. 20) and normal people (0. 33 ± 0. 21)(p>0.05).There was significant difference between patients with breast cancer after (0. 60 ± 0. 42) and before operation, normal people respectively (p<0.01).The expression of apoptosis(Apo) in peripheral blood of patients with breast cancer after operation (0. 40 ±0. 20) was very lower than that of patients before operation(2. 14±0. 76) (p<0. 01), and was almost equal to that of normal people (0. 45 ± 0. 13) (p>0.05). There was significant difference between patients with breast cancer( before operation: 2. 48 ± 0. 91, after operation: 2. 55±0. 80) and normal people(4. 20 ± 0. 82) in pl6 protein expression (p<0. 01). The expression of pl6 protein in patients of breast cancer with lymph nodal non-metastases (1.99 ±0.44) was higher than that of patients with lymph nodal metastases (0. 45 ± 0. 40) (p<0. 01). The expression of pl6 protein in Ⅰ and Ⅱ stages (1.00 ± 0.08) of breast cancer patients was higher than that of Ⅲ and Ⅳ stages (0.84±0.09) (p<0. 01). There was significant difference statistically between patients with breast cancer( before operation: 1.25 ± 0.93, after operation: 1.16 ±0.52) and normal people(0. 56 ± 0. 30) in peripheral blood of cyclin D1 protein expression (p<0. 05). The expression of cyclin D1 protein in patients of breast cancer with lymph nodal non-metastases (1. 26 ± 1. 05) was lower than that 6f patients with lymph nodal metastases (2. 32±1. 36) (p<0. 05). The expression of cyclin Dl protein in Ⅰ and Ⅱ stages(1. 00±0. 02) of breast cancer patients was lower than that of Ⅲ and Ⅳ stages (1. 28 ± 0.14) (p<0. 01). D1 was associated with SPF and pl6 statistically inperipheral blood of patients with breast cancer before operation, the correlationcoefficient was 0.73 and 0.40, respectively (p<0.01), and there was significant relationship between the last two, the correlation coefficient was 0.42(p<0.01).Conclusion: 1, The expression of pl6 and cyclin Dl protein in peripheral blood of patients with breast cancer was associated with lymph nodal metastases and clinical stages, there was not any significant difference statistically between patients with breast cancer after and before operation of the expression of p16 and cyclin D1 protein.2, The rate of SPF was higher and Apo was lower after operation than...
