Aim To investigate the effect of adrenaline on expression of lipid traffic-related genes and to identify its possible mechanism mediated by adrenoceptor.Methods THP-1-derived macrophages were incubated with various concentrations (1pM to 1 μM) of adrenaline for 24 hours.Then TGF- β 1,CD36,ABCA1,SR-AI and Caveolin-1 mRNA expression were determined by RT-PCR. Next, cells were preincubated for an hour with an alphal-adrenoceptor antagonist prazosin(100 pM to 1μM), an alpha2-adrenoceptor antagonist yohimbine(1nM to10μM), a betal- adrenoceptor antagonist metoprolol(10nM to 100μM) and a beta2- adrenoceptor antagonist butoxamine(2.5 nM to 25 μM) , respectively, before treatment with 1μM adrenaline for 24 hours . Then these genes in THP-1-derived macrophages were analyzed by RT-PCR.Results After THP-1-derived macrophages were exposed to the different concentrations (1pM to 1μM) of adrenaline for 24 hours,RT-PCR analysis showed that, physiological concentrations(1pM to 100pM) and stressful condition(lnM to 10nM) of adrenaline had no significant effect on these genes expression (P>0.05). Meanwhile, compared with control (without adrenaline), adrenaline of higher concentrations(100nM , 1μM) significantly decreased TGF- β 1mRNA expression by17.2% and 14.7% (P<0.05), ABCA1 mRNA expression by 9.5% and 10.4% (P<0.05)and significantly increased SR-AI mRNA expression by 13.5%and 11.7%...
