Objective: Colon cancer is one of the common cancer in china, it's morbidity is in the forth place in gastrointestinal cancer. There are about 400 thousand men and 380 thousand women get this disease each year, in resent years, more and more people suffering from this disease in china .The main treatments for colon cancer are surgery, chemotherapy, radiotherapy and gene therapy. We have found the limitation of surgery. Gene therapy is also limited for it's high expend. It is important to find a better method to direct the treatment of colon cancer, early diagnosis and synthetically therapy are important to colon cancer. IL-6, IL-10, IL-12 and IFN- γ are important cell factor in our body. In this experiment, through abserve the level of IL-6, IL-10, IL-12 and IFN- γ mRNA in peripheral blood monoeuclear cell of colon cancer ,We can find which kind of treatment is the best way for patient's immune state , thus we can afford better treatment to colon cancer.Method: We use the technology of reverse transcription polymerase chain reaction (RT-PCR) in this experiment, (1)Through abserve the level of IL-6 , IL-10, IL-12 and IFN- γ mRNA in peripheral blood monoeuclear cell of colon cancer, we can know the different between colon cancer patient and normal people. (2) Through abserve the level of IL-6, IL-10, IL-12 and IFN- γ mRNA in peripheral blood monoeuclear cell before and after chemotherapy, we can know the effect of chemotherapy. (3) Through abserve the level of IL-6, IL-10, IL-12 and IFN- γmRNA in peripheral blood monoeuclear cell in different group (A and B), we can know the different curative effect between biotherapy plus chemotherapy group and chemotherapy group only.Results: (l)We found that the level of IL-6, IL-10 and IL-12mRNA in peripheral blood monoeuclear cell of colon cancer patient is higher than normal people (PO.05 ) , The express of IFN- y mRNA in peripheral blood monoeuclear cell of colon cancer patient is negative .(2) The level of IL-6, IL-10 and IL-12mRNA in peripheral blood monoeuclear cell of colon cancer patient was higher than control group before chemotherapy and reduced after chemotherapy(PO.05) .(3) The level of IFN- Y mRNA in peripheral blood monoeuclear cell of colon cancer patient was lower than control group before chemotherapy and rose after chemotherapy. (4)Biotherapy combined with chemotherapy group can reduce the level of IL-6, IL-10 and IL-12 more effective than chemotherapy group only(PO.05), The level of IFN- Y mRNA in peripheral blood monoeuclear cell of colon cancer patient rose from the second cycle (P=0.043). The level is the highest in the third cycle(P=0.030).Conclusion: (Dthe level of IL-6, IL-10 and IL-12mRNA in peripheral blood monoeuclear cell of colon cancer patient is higher than normal people , The level of IFN- Y mRNA in peripheral blood monoeuclear cell of colon cancer patient is lower than normal people , Thl cell drift to Th2 cell, contribute to cancer's development. (2)Chemotherapy can reduce the expression of IL-6, IL-10 and IL-12 in colon cancer, rise the expression of IFN- y , enhance the patient's immune state. (3) Biotherapy combined with chemotherapy can reduce the expression of IL-6, IL-10 and IL-12 in colon cancer greatly, rise the expression of IFN- Y deeply. Thus we can improve the immune state of colon cancer patient, constrict the grow of cancer, improve the patient's prognosis , help the patient recover from illness.(4) Biotherapy should be applied for a long time , it's side effect is small ,so it can contribute to patient recovery greatly.
