Objective: Gastric carcinoma is considered to be one of the most common malignant tumor in China. Annual over 260,000 patients died of gastric carcinoma, accountable for the 20 percent of all cancer death.So gastric carcinoma has been one of the leading causes of cancer mortality every year. The treatments of gastric carcinoma mainly include surgery, chemotherapy, radiotherapy and biotherapy. Chemotherapy is one of the main therapeutic means for gastric carcinoma. Cellular immunity is closely related to occurrence and development of tumors. Some studies show there is a drift from Thl to Th2 in tumor cells. No enough data is available about the effect of chemotherapy on cellular immunity and the levels of Thl and Th2 cytokines in patients of malignant tumors. Moreover, the reported research about this aspect is controversial. This study is to investigate the gene expression of Thl and Th2 cytokines in peripheral blood mononuclear cells (PBMCs) of patients with gastric carcinoma before and after chemotherapy; to explore the immune status of patients with gastric carcinoma; and to evaluate the effect of chemotherapy and biotherapy on patients' cellular immunity; and to find the relationship with development and prognosis of tumor.Method: We use the technique of reverse transcription polymerase chain reaction (RT-PCR) in this experiment, to detect the levels of Thl and Th2 mRNA in peripheral blood mononuclear cells (PBMCs) of patients with gastric carcinoma or benign diseases and normal people, and analyze the relevance between the clinical stage and histological differentiation of gastric carcinoma and the levels of Thl and Th2 mRNA.
