Objective: To explore the change of sputum and serum concentration of IL-13,TNF-αand IFN-γin the acute exacerbations of Asthma and Chronic Obstructive PulmonaryDisease(COPD), and to compare concentrations of difference of thesecytokines in sputum and serum. To analysis of the characteristic inflammatory cellin induced sputum , and to compare the relationship between these cytokines and thecharacteristic inflammatory cell in sputum, and to compare the relationship betweenthese cytokines and FEV1%pre.Methods: Sputum and serologic analysis through enzyme-linked immunolosorbentassay(ELISE) were used to detect the sputum and serum concentration of IL-13,TNF-α,IFN-γin 20 healthy control subjects , 21 patients with acute exacerbationof Asthma ,23 patients with acute exacerbation of COPD. At the same time,inducedsputum was scribbled to count inflammatory cell, and compare inflammatory cellin the three groups. To analysis correlation of cytokines IL-13,TNF-α,IFN-γandthe percentage of sputum inflammatory cell , to analysis correlation of cytokines IL-13,TNF-α,IFN-γwith the percentage of the first second forced expiratory volu-me/predicted value (FEV1%pre).Result: 1.The differences of the eosinophil(EOS)/leukocyte%(EOS/Leu%) in sputum inasthma group(14.6±5.8) and in control group(1.6±0.4),in COPD group(1.8±0.9) weresignificant(P= 0.000,P= 0.002).The EOS/Leu% was significantly inversely correlated withthe percentage of the first second forced expiratory volume/predicted value (FEV1%pre) inasthma group(r =-0.605,P= 0.004). 2.The differences of the neutrophil/leukocyte%(Neu/Leu%) in COPD group(61±6) and in control group(24±5), in asthma group(23±7) were significant(P= 0.000,P= 0.000).The Neu/Leu% was significantly inverselycorrelated with FEV1%pre in COPD group(r = -0.842,P = 0.000).3. The sputum co-ncentration of LogIL-13 in patients with Asthma group(2.09±0.25) were much hig-her than in the control group(1.71±0.19) and in the COPDgroup(1.74±0.28)(P =0.000,P = 0.000),while there was no difference between the controls groupand COPD group(p>0.05). The serum concentration of IL-13 in patients with Asthma groupwere much higher than in the control group and in the COPD group(P<0.05).4.The sputum concentration of IL-13 was significanty inversely correlated with FEV1%pre in Asthma group (r = -0.752, P = 0.000) and the sputum concentration ofIL-13 was significantly positively correlated with EOS/Leu% in Asthma group (r =0.832,P = 0.000). 5. The sputum concentration of TNF-αin patients with Asthmagroup(53.38±13.28)and COPD group(63.19±12.30) were much higher than the cont-rols group(15.20±4.99) (P = 0.000,P = 0.000),while there was no difference betw-een the Asthma group and COPD group (P>0.05).6. The sputum con-centration ofIFN-γin patients with Asthma group (21.90±5.16) and COPD group(19.79±5.31)were much higher than the controls group (12.47±4.65) (P = 0.000,P = 0.000)),while there was no difference between the Asthma group and COPD group(P>0.05).7. The serum concentration of TNF-α,IFN-γin patients with As-thma group andCOPD group were much higher than in the controls group(P<0.05).The sputumconcentration of TNF-α,IFN-γwere much higher than in the serum concentrationof TNF-α,IFN-γin Asthmagroup and COPD group(P<0.05).Conclusion:1.Activation of eosinophil, more releasing of IL-13,TNF-α,IFN-γallparticulate in the airway inflammation in exacerbations Asthma. It is suggested theIL-13 might be involved in the pathogenesis of asthma via eosinophil regulation.2.Activation of neutrophil, more releasing of TNF-αand IFN-γall particulate in theairway inflammation in exacerbations COPD.
