Objective: To investigate the relation between induced sputum cell counts and lung function in patients hospitalized for chronic obstructive pulmonary disease.Methods:Applicated 4.5% hypertonic saline as an inducer.The patients preventive inhaled 200μg salbutamol aerosol before tested. 10min after this, inhaled 4.5% hypertonic saline for 15min-20min with 4mL/min, and then by cold water rinse, blow your nose. Told patients deep cough sputum to dish about 3mL (at least greater than 0.5mL), and then added 4 times the volume of sputum 0.1% of DTT, watered bath at 37℃for 20min mix full oscillation dissociation, and filtered with the objective aperture of 200 nylon gauze, to 2000r/min, centrifuged 10min (centrifugal radius of 10cm), discarded supernatant, precipitation part of the application of Wright stain was smeared and dried, after this, did cell classification under the optical microscope. Classificated 200 white blood cell and calculated the percentage, including eosinophils (EOS), neutrophil (N), and macrophages (M). In the light microscope, the squamous cells occupied more than 20% in nucleated cells as the proportion of unsatisfactory sputum, and they should be excluded. The group of patients with lung function and inflammatory markers was in check. Lung function indicators included: total expiratory volume in one second percentage of predicted value (FEV1%pre), expiratory volume in one second vital capacity ratio of occupied (FEV1/FVC) and forced mid-expiratory flow (FEF25-75%); inflammatory markers included: high sensitivity C-reactive protein (hs-CRP), procalcitonin (PCT). Using SPSS17.0 statistical software, analyzed the associateion of cell counts of induced sputum in chronic obstructive pulmonary disease with lung function.Results:1) Induced sputum neutrophils in COPD patients hospitalized increased significantly comparingt with healthy individuals (P<0.05), and percentage of eosinophils in COPD patients hospitalized than the control group, was significantly higher(P<0.05). Macrophages was mainly in induced sputum of healthy people(P<0.05);2) hs-CRP and PCT in COPD patients hospitalized with COPD were higher than normal health (P<0.05);3) Induced sputum neutrophils of COPD hospitalized were negatively correlating with FEV1%pre(r=-0.481, P<0.05), and negative correlation was also found with FEV1/FVC (r=-0.583, P<0.05).Conclusion: Counts of neutrophils of induced sputum in patients with COPD hospitalized correlates with lung function, suggesting that neutrophils in the pathogenesis of COPD has an important role.
