OBJECTIVE:To elucidate the effect and possible mechanism of polysaccharideextracted from Aloe barbadensis Miller on wound healing and provide thepharmacological foundation of drug exploitation about wound healing of Aloe.METHODS:Polysaccharide was isolated and purified from Aloe barbadensis Millerand study its effect on fibroblast and epidermal cell cultivated in vitro then prepare theexternal use ointment and establish the quality control method. At last, study the effectand possible mechanism of ointment used on superficialⅡscald.RESULTS:A polysaccharide, which was mainly β(1→4)-D-acemannan, was isolatedfrom the fresh Aloe barbadensis Miller leaf. Cell cultivation in vitro results showed thatthe polysaccharide can promote the fibroblast and EC growth when Conc. higher than100μg/ml. The promotion effect depended on the AP Conc. in culture fluid, at the sametime, the results showed no distinguished difference. Then we successfully prepared theexternal use ointment involved AP and established the quality control method. Animalexperiment showed that the ointment could speed up the superficialⅡscald healing rate ofmice significantly without observating any side effects on liver and kidney.CONCLUSION:AP extracted from the fresh Aloe barbadensis Miller not only canpromote the fibroblast and EC growth but also can speed up the superficialⅡscaldhealing rate of mice. At the same time, the study proved the safety of AP. In a word, thestudy results show that AP may be exploitated to some new drugs about promoting woundhealing with optimistic perspective.
