Purpose: Hyperlipemia model and atherosclerosis model were established by feeding forage containing 2% cholesterol.The purpose of the experiment is to compare the effect of the combination of aspirin and Simvastatin with Simvastatin on atherosclerosis and discuss the mechanism. Method: 24 female SD rat were randomly distributed into 2 guoups.One group(NC group) has six rats which were fed with standard forage; the hyperlipemia model group(H group) has eighteen rats which were fed with forage containing 2% cholesterol. After four weeks,H group was randomly distributed into there guoups.The first group(HL组) was fed without drug;The second group (S组) was infused with simvastatin (10mg/kg*d);The thire group (A+S组) was infused with simvastatin (10mg/kg*d) and aspirin(25mg/kg*d);Every group had six rats.All rats were fed for 8 weeks continuously to establish atherosclerosis model.Respectively at the end of 4 weeks and 12 weeks,all rats' blood fat were measured by the radiation and immunity method.The area of the plaque and endomembrane and the perimeter of the aorta and the ratio of plaque/endomembrane in the area were measured by quantitative pathology.The atherosclerotic change of the aorta was observed by optical microscope.The quantity of advanced glycation end products (AGEs)in thoracic aorta and abdominal aorta was measured by fluorospectrophotometer. Result: Hyperlipemia model and atherosclerosis model were confirmed by according to pathology and blood fat results.There was no significant difference on blood fat between A+S group and S group.Observation by naked eye: The aorta endomembrane of NC group was of smooth and thinness without plaque; The aorta endomembrane of HL group was of rough and thick with fusion of plaque. The aorta endomembrane of A+S group and S group was of smooth with sporadic plaque.The area of plaque and the ratio of plaque /endomembrane in area significantly decreased in A+S group than in HL group(P<0.05).The difference between A+S group and S group was not significant,but the tendency decreased.Observation by optical microscope :NC group kept complete aorta endothelial structure without calcification.HL group hadn't integrated aorta endothelial structure and had significantly calcified tunica media with proliferation of collagen.Both A+S group and S group had integrated aorta endothelial structure.Thecalcification of tunic media on A+S group and S group was lighter than on HL group.The calcification of tunic media on A+S group was lighter than on S group.The comparison of AGEs content: The AGEs content on HL group was significantly higher than NC group(P<0.05). The AGEs content on A+S group was significantly fewer than S group(P<0.05). Conclusion: Rats fed with high lipoid-diet showed higher numerical value of blood fat and thickening endomembrane and calcified tunica media.Aspirin didn't significantly change blood fat of rats.The combination of simvastatin and aspirin significantly decreased AGEs content and calcification of aorta and released the advancement of atherosclerosis.
