Objective : To investigate the expression of survivin in osteosarcoma,osteochondroma and normal osseous tissue, and its correlation with clinic pathologiccharacteristics, and its relationship and clinical significance with expression ofP-glycoprotein, Bcl-2 and VEGF in osteosarcoma respectively.Methods:Thirty-eight specimens (gender distribution:23 males and 15 females)were obtained from the patients with osteosarcoma at the Department of Orthopaedicssurgery First Affiliated Hospital of Jiangxi medical college during the period from1998 to 2003. All patients received neither chemotherapy nor radiation therapy beforebiopsy..Using immunohistochemistry(S-P) method, the expression of survivin inosteosarcoma, osteochondroma and normal osseous tissue, and the expression ofP-glycoprotein, Bcl-2 and VEGF in osteosarcoma were detected.Results:Survivin positive expression rate was 63.17% in osteosarcoma, and noexpression of survivin was detected in osteochondroma and normal osseous tissue.The positive expression rate of survivin was significantly associated with Ennekingclinical stages and histological typing (WHO), but no relationship among survivinexpression and age, sex and tumor location. The positive expression rate ofP-glycoprotein, Bcl-2 protein and VEGF were 47.37%, 55.26% and 47.37%respectively. There was a significant correlation between survivin and p-glycoprotein,between survivin and bcl-2, and between survivin and VEGF.Conclusions:1) Survivin is over-expression in osteosarcoma. It is probablyinvolved in the etiology of osteosarcoma and maybe an important reference marker inevaluating the prognosis of osteosarcoma. 2) Detecting early the expression ofsurvivin and p-glycoprotein to patients with osteosarcoma is helpful to evaluate thechemotherapy sensitivity and clinical multidrug resistance. 3) The expression ofsurvivin was closely correlated to that of Bcl-2 and they may play a synergic role inthe course of anti-apoptosis. 4) Survivin and VEGF expressions, both contributing tothe tumor neovascularization, may be helpful in the understanding of intra-tumoralangiogenetic mechanism in the future.
