Objectives: To study the potential molecular mechanism of the chemosensitive difference between human ovarian epithelial and germ cell tumor by testing the expression of p53(mutation),Bcl-2,Topo Ⅱα in paraffin embedded tissue from human ovarian tumor.Methods: 1)Tissue microarray was manually made with the tissue from human ovarain epithelial(n=53) and germ cell(n=25) tumor. 2) The expression of p53(mutation),Bcl-2 and Topo Ⅱα in the tissue microarray was tested by immunohistochemical staining. 3)A11 data was analysis by rank sum test.Results: The positive expression rate of p53(mutation),Bcl-2 and Topo Ⅱα was 56.60%,24.53% and 49.06% in the sample from ovarian epithelial tumor, and 28.00%,32.00% and 44.00% in the sample from ovarian germ cell tumor. The expression of p53(mutation) was higher in the epithelial tumor than that in the germ cell tumor, whereas the expression of Bcl-2 and Topo Ⅱα had no difference between the two groups.Conclusion: A lot of factors are associated with the difference of chemosensitivity between human ovarian tumors.Our results suggest that p53 may have important molecular effect on the difference of chemosensitivity. Tissue microarray is efficient to test the ovarian tumor tissue by immunohistochemical staining.
