Objective To observe the anticancer effect in vitro on human lung cancer cell lines NCI-H460 and A549 using anti-hnRNP B1 monoclonal antibody combined with DDP. The final purpose of this experiment is to provide some theoretical evidences for the biologic therapy of lung cancer. Methods DDP was combined with anti-hnRNP B1 monoclonal antibody to produce the targeting drug. The human large cell lung cancer cell line NCI-H460 and human adenocarcinoma lung cancer cell line A549 were cultured with culture medium in vitro. The human lung cancer cells were divided into four groups randomized. They are blank group, DDP group, anti-hnRNP B1 monoclonal antibody alone, and the targeting drug groups. The tumor cells were treated with different dose of the drugs above in 12, 24, 48 hours. The inhibition rate was determined by the number of cells which intake 3H-TdR. The apoptosis was observed by TUNEL. And the cell cycle was determined by flow cytometry.Results (1) With the treatment of the drugs above, the number of the cells which intake 3H-TdR decreased. 1) In NCI-H460, the cell number in DDP group is 743±31 in 48hours. In hnRNP Bi group the number is 686±17. In hnRNP Bi-DDP group the number is 586±25. In A549, the cell number in DDP group is 654±41. In hnRNP Bi group the number is 607±16. In hnRNP Bi-DDP group the number is 531±27. There is a obviously significant difference between the hnRNP B,-DDP group and DDP, hnRNP Bi group (P < 0.01). It indicates that the hnRNP Bi-DDP group has a better inhibition to the lung cancer cells. 2) In NCI-H460, the cell number in blank group is 1370±24. In DDP group the numbers are 1009±23, 897±31 and 743±31 in 12, 24, 48hours. In hnRNP Bi-DDP group the numbers are 768±13, 579±20 and 446±32 in 12, 24, 48 hours. In A549, the cell number in blank group is 1300±28. In DDP group the numbers are 975±2, 763±17 and 654±41 in 12, 24, 48hours. In hnRNP Bi-DDP group the numbers are 763±45, 618±20 and 432±19 in 12, 24, 48 hours. And with the increase of time, the inhibition rate increased, which indicates that the inhibition rate was time dependent. 3) In NCI-H460, the cell numbers in 1.5/^g/ml, 3/Wg/ml and 6/jg/ml group are 735±19, 586±25 and 446±32 in 48 hours. In A549, the cell numbers in 1.5/xg/ml, 3/ig/ml and 6/ig/ml group are 656±30, 531±27 and 432±19 in 48 hours. They all suggest that the inhibition rate is dose dependent. A obviously significant difference could be observed between the hnRNP Bi-DDP group and DDP, hnRNP Bi group (P < 0.01). (2) The apoptosis body was seen after the treatment of the hnRNP B,-DDP. (3) After the treatment of the hnRNP Bi-DDP, the cell cycle of NCI-H460 was blocked at G0/Gi phase, which accounts for 56.4%, and the number of the cells increased at this phase.However, the number of the cells decreased at G2/M phase, which accountsfor 35.8%. At the same time the cell cycle of A549 was also blocked at G0/G1phase, which accounts for 53.9%, while G2/M phase accounts for 43.2%.There is a obviously significant difference between the hnRNP Bi-DDPgroup and DDP, hnRNP Bi group (P < 0.01).Conclusion (1) The anti-hnRNP Bi monoclonal antibody is highly specificto the lung cancer cells and can inhibit the growth of the human lung cancercells. (2) The targeting drug made from anti-hnRNP Bi monoclonal antibodyand DDP has the targeting anticancer effect in vitro on human lung cancercells.
