Objective : To observe the effect of shengmai injection (SMI) on the myocardium injury by adriamycin (ADM) in rats and the mechanism involved , and compare the protection effect of the SMI group â…  with the SMI group â…¡ . Methods : The molders of ADM-induced myocardial damage of rat was used . 40 Wistar rats were randomly divided into four groups . In the normal group , normal saline was intraperitonial (ip) injected at a dose of 5ml/kg once a day for 12 days ; the ADM group were given ip injection of ADM at a dose of 5ml/kg (0.6mg/ml) from the 4 th day , on every other day for 5 times and normal saline at a dose of 5ml/kg once a day for 12 days ; The SMI group â…  were ip injection of ADM at a dose of 5ml/kg from the 4 th day , on every other day for 5 times and SMI at a dole of 5ml/kg once a day for 12 days ; The SMI group â…¡ were given ip injection of ADM at a dose of 5ml/kg from the 4 th day , on every other day foe 5times and SMI at a dose of 5ml/kg from the 4 th day , once a day for 9 days . The activities of superoxidized dismutase (SOD) , succinate dehydrogenase (SDH) ,Ca2+-ATPase and content of malondialdehyde (MDA) in the myocardial mitochondria were observed . The rat's myocardial histopathological observation was thesame time .Results : In the ADM group,the content of MDA increased and the activity of SOD , SDH , Ca2+-ATPase decreased significantly (P<0.0l) .Compared with those in the ADM group , the content of MDA decreased (P<0.0l) in the SMI group I and the activities of SOD ^ SDH > Ca2+-ATPase increased significantly (P<0.05); whereas the SMI group II only showed significant decreased in the content of MDA (P<0.05). besides the activity of SOD ,There was a significant difference between the SMI group I and II (P<0.05) .Compared the SMI group I with the normal group had no significant difference (P>0.05) . Pathological examination and electron microscopy observation demonstrated that the cardiac muscular tissue and ultrastructures were harmed at different levels in the every group except the normal group . Compared with the ADM group , the pathologic change of myocardium was significant reduced in the SMI groups . Conclusion : The SMI has protective effect on cardiotoxicity induced by ADM . The mechanisms may depend on the effect of SMI on scavenging oxygen free radials in rat heart , inhibiting lipid peroxidation , decreasing calcium overload and ameliorating myocardial energy metabolish . Meanwhile , treated with SMI on different time and doses after the cardiac muscular tissue injury caused by adriamycin , which would be researched further .
