In recent years, in experimental research and clinical observation, peoplefound that the function metabolize obstructed and construction destructed ofthe cell have became more serious after the tissues have been recovered theblood.Though the tissue and organ of ischemia all can occur ischemicreperfusion injury,but the tissue and organ which have highly volume ofdemand of oxygen, such as heart,brain and so on,will occur ischemicpeperfusion injury easily.So the phenomenon has been named ischemiapeperfusion injury. The first knowledge about ischemia reperfusion injury ismyocardial reperfusion injury(MRI).The flare up mechanism of MRI which isknown are oxygen free radicals(OFR) and calcium overload and so on. OFRand lipid peroxidation which is related to OFR are the start factot ofmyocardial injury. The calcium overload in the cell is the common link of theinjury. The energy exhausted, oxidative phosphrylation disjointed, the releaseof enzyme from the cell and the destruction of the cell membrane are thecommon result of MRI. People have adopted methods (such as get rid of OFR,lighten calcium overload ect) in order to protect myocardium. Even thoughnow people have been gone on a lot of experimental and clinical studies, butwe have no efficacious prophylaxis and treatment method of MRI. Themodern pharmacodynamics study proved :the efficacious composition of someof the tradition Chinese medicine have the effection of protect ischemiamyocardium, get rid of OFR . Shengmai Injection(SMI) is the high efficiencyextract of the tradition Chinese medicine Shengmai-san by modernpharmaceutical technology.In clinical ,SMI is used on Coronary heart desease,Cardiac insufficiency,various shocks ,especially cardiogenic shock.This studyis to observe the protective effect of SMI on myocardium of rabbit withischemia reperfusion injury and also discuss the function mechanism of SMItreating MRI.In experimental research, we divided 30 healthy rabbits into 3 groups,including no ischemia group (I group), ischemia reperfusion group (II group)and ischemia reperfusion SMI group (III group). There were 10 rabbits inevery group. Accordingto the ligate-loosen way copy MRI rabbits. When theMRI model have been accomplished, we given physiology salt solution to IIgroup 2ml/kg before the left anterior destending branch of coronary arterywas igated, and then given physiology salt solution to II group 5 ml/kg/hcontinuous with the micro-pump when the blood were recovered. In III group,we used SMI instead of physiology salt solution. We got the blood after theexperimental finished. The blood were given to examine the content of bloodserum myocardial enzyme, including Creatinine phosphokinaseCPK,CK-MB.The rabbit's hearts were sent to make electro-microscope sepciment and thento observe ultrastructure of myocardium. The activity of superoxide dismutase(SOD) and the contentration of malondialdehyde (MDA) were assayed. Allof them would be compared in order to see whether they had the dramaticaldifference.The exerperimantal research showed the following result : 1. SMI couldget rid of OFR and prevent cell lipid peroxidation. MDA contention of IIgroup was higher than that of I group. While SOD activity in II group waslower. SOD activity of III group was higher than that of II group. While MDAcontention in III group was lower, have dramatically different hrough thestatistical analysis, P<0.01.2.SMI could protect the myocardial cell membrane.In the exerperimental research, SMI could decrease the release of myocardialenzyme in III group, have dramatically different through the statistical analysis,P<0.01. 3. SMI cam improve ultrastructure of myocardium of ischemiareperfusion injury.
