Objective: To study the differences in aberration of pl6 and cyclinDl gene in patients with non-small cell lung cancer (NSCLC) between Xuanwei county and Kunming city. To explore the role of pl6 and cyclinDl gene in the genesis and development of NSCLC.Methods: 45 cases of NSCLC from Xuanwei county and Kunming city respectively, 18 cases of pneumatocele or pulmonary hemorrhage as normal control were collected. Immunohistochemical staining was used to analyze the expression of pl6 and cyclinDl, PCR-SSCP and direct sequencing were used to analyze pl6 exon2 deletion, point mutation and cyclinDl (CCND1) G870A polymorphism in the samples of paraffin-embedded tissues in lung from these 108 case.Results: P16 protein total loss expression incidence was 41. 7%, while Xuanwei , Kunming and control group was 57.8% , 37.8% , 11.1% respectively. Comparing the loss expression of pl6 protein in three groups, there were statistical significant differences (P<0. 05) :cases of stage I, adenocarcinoma and stage T2 in Xuanwei group were higher than that in Kunming group respectively; Xuanwei group and Kungming group was higher than control group respectively.Total cyclinDl protein expression incidence was 51. 9%, while Xuanwei, Kunming and control group was 55. 6%, 60. 0%, 22. 2% respectively. There were statistical significant differences in expression incidence of cyclinDl (P<0. 05): stage I in Xuanwei group was higher than that in Kunming group; but adenocarcinoma in Xuanwei group were lower than that in Kunming group;the expression incidence of cyclinDl in Xuanwei and Kungming group was higher than that in control group respectively.Among 90 NSCLC cases, 26 cases deletion and 15 cases point mutation of pl6 exon2 had been found. Incidence of the deletion and mutation in Xuanwei group was 33. 3%, 22. 2% respectively; while Kunming group was 24. 4%, 11.1%; two cases of control group had deleted and mutation was not found. There were statistical significant differences in the deletion and mutation incidence of pl6 exon2 (/K0. 05): Xuanwei group was higher than control group; adenocarcinoma in Xuanwei was higher than that in Kunming. Kunimng group was no difference with control group. The total deletion and mutation incidence of pl6 exon2 in Xuanwei and Kunming group was no difference. 13 mutational site locating in C or G had been found.AA, AG, GG of G870A genetype probability in Xuanwei group was 46. 7%, 42. 2%, 11.1%; Kungming group was 24. 4%, 44. 4%, 31.1%; control group was 22. 2%, 38.9%, 38. 9%. AA of G870A genetype total probability in Xuanwei group was higher than that in Kunming group(K0. 05); which in stage T2 and with lymph node metastasis in Xuanwei group was higher than that in Kunming group (K0. 05).Conclusions:P16 protein loss expression in cases of stage I , adenocarcinoma and stage T2 in Xuanwei group had difference in Xuanwei and Kunming; cyclinDl protein overexpression in stage I and adenocarcinoma had difference in Xuanwei and Kunming,- The incidence of pl6 gene inactivation, deletion and mutation had difference in adenocarcinoma in two region; 13 mutational site locating in C or G had been found; AA of G870A genetype in Xuanwei group were higher than Kunming group.
