It has been proved that the immune function of host with hepato-cellular carcinoma(HCC) is decreased to some extent,and the cellular responses of the host to the tumors could restrict the development and growth of HCC.How to improve the host's cellular responses is a key step to curb the cancer growth in a researcher's point of view.Skin allograft could stimulate the host's acute immune rejection towards the graft. Because transplant antigen and tumor-cells both express human leukocyte antigen(HLA), and the graft skin and the tumor tissue are both foreign objects to the host's immune system, therefore the rejection immune responses of the host to the allograft is also apply to the tumor tissues.The aims of our project are:1.to prove that skin allograft in Kunming mice with H22 HCC cell could induce the host's immune response to tumors.Concretely,we tried to observe the difference between the skin allograft groups compared to the control groups by observing the weight difference as well as tumor cell apoptosis by TUNEL staining method between the experiment group and the control groups.2.to observe the peripheral blood lymphocyte(PBL)difference between the allograft group and the controls.Methods:The Kunming mice with hepato-cellular carcinoma(HCC)were used as model and divided into 3 groups.After subcutaneously injection of H22 cell suspension,the experimental group(15 mice)received skin allograft from C57/BL6,and the other two groups(15 mice versus 10 mice)receiving the skin grafts from same line mice or no grafts both as controls.We also observed the peripheral blood lymphocyte subsets(CD3+T cell,CD4+T cell,CD8+T cell,CD19+B cell and NK1.1+NK cell)difference between these groups.Results:A slower tumor growth rate was found in the experimental group(P<0.05),and we also observed more apoptosis tumor cells in the experiment group compared with the controls(P<0.05),but there is no differences between different groups on peripheral blood lymphocyte subgroups(P>0.05).Conclusion:The skin allografi could inhibit the H22 growth in Kunming mice probably by more tumor cell apoptosis mechanisms,which may provide some new ideas in exploring the clinical regime in patients with HCC in the future.
