Objective Paraquat, as a herbicide, is polarity compound and easily soluble in water. For this properties, there is a major obstacle to separate it from matrix in realistic cases. The ion-pairing solid-phase extraction was used in this paper to carry out the extraction and clean-up simultaneity. A HPLC-DAD and HPCE-DAD method were developed for determination and identification of Paraquat respectively. The advantages of the method about extraction, the separation from biological samples and the determination of paraquate were testified by analysis of the samples of poisoning rabbits and plants experiments. Method the biological material were digested with enzyme, Then the digest solution were extraction by ion-pairing solid-phase extraction column, quantitative analysis were carried out with HPLC, and identification with HPCE-DAD and TLC. Results The recoveries of Paraquat in biological sample were about 81%~94% and the limits of detection were lng·ml~-1. Conclusion the advantages of the method with ion-pairing solid-phase extraction for Paraquat in biological sample, water and poisoning plants are higher selective, extraction solution are cleaner and have better reproducibility. HPLC method is accurate, sensitivity and good reproducibility. TLC method is rapid and useful for analysis of Paraquate in poisoning plants and CE method has filled the gaps in this field in our country. The research provides a scientific reference and the analysis method for the cases of poisoning of human, animal and plants.
