Time-resolved fluorescence immunoassay (TRFIA) is a new immunoassay technology established in 1970s. Lanthanide rare-earth ion chelates have their unique fluorescence properties: high quantum efficiency, large Stokes'shift between excitation and emission,narrow emission band, long decay time and very sharp emission peak. These can make that sample is determined in very narrow wavelength though adjusted instrument, and decrease disturbance of all background signals. Using the chelate of rare-earth ion labeling Ag or Ab, the high sensitivity of rare-earth ion detection can be obtained with time-resolved fluorometry, using befitting delayed time and measuring wave. The disturbance of background signal can be eliminated effectively and increase detected sensitivity. TRFIA have some advantage as follows. High sensitivity, strong specificity, good stability, conveniently prepare of the label, long store time, non-radioactive pollution, measure repetitiveness to be fine, simple operation procedure, wide linearity range, extensive applied range. TRFIA becomes a new milestone in labeling immunoassay after radioimmunoassay (RIA). TRFIA is the most developing future biochemical ultra-trace detection method in ligand combination analysis at present.
