| In this experiment, we investigate the effects of this traditional Chinese medicine on osteogenic and proliferation differentiation of bone marrow stromal cells in vitro. Different concentrations of ethanol extract of Gu-Sui-Bu were added to rabbit marrow stromal cells culture.The effect on osteoblastic differentiation was examined by reserved phase-contrast microscope, histochemistry stains. The effect on proliferation was examined by MTT. In the same time, the proliferation and osteoblastic differentiation of these cells cultured by pure medium were examined in same methods. The effective concentrations of Gu-Sui-Bu on osteoblastic differentiation of were 2mg/ml 0.2mg/ml and 0.02mg/ml.The addition of 2mg/ml and 0.2mg/ml respectively to bone marrow stromal cells culture partly expressed osteoblastic phenotype. Firstly these cells induced began to differentiation or transform, then proliferated and secreted, ultimately came into being mineralized matrices in vitro. Histochemical studies showed these cells could secrete alkaline phosphatase and collagenâ… ,mineralized matrix to form calcific nodules so much as bone-like tissue. Positive rate of alkaline phosphatase stains preponderated over 30%. The analysis of Image-Pro Plus of alkaline phosphatase and collagenâ… stains,and the result of count of calcific nodules demonstrated that the effect of the two concentrations of Gu-Sui-Bu were more obvious than pure medium, while there were no difference between the two concentrations. The analysis of MTT demonstrated that the preferable effect of concentrations is 0.02mg/ml. In this study, we demonstrated that Gu-Sui-Bu contains ingredients that can affect the proliferation and osteogenic differentiation of bone marrow stromal cells. Perhaps the effect of Gu-Sui-Bu is a kind of cytology mechanisms of the Chinese medicine functions of promoting the concrescence of bone fracture and preventing osteoporosis.
