Methods such as GC-MS, 13CNMR, 1HNMR, 2D1HNMR etc were used to analyze the structure of polysaccharide from Semen Cassiae seeds (PC). The results showed that the(l-4)linked mannose back bone with(l-6) attachment of galactose clarified the structure [4) -gal-(1-6)-mannose( 1 -4)-(mannose)5-( 1 -]nThree-line crossbred (Duroc × (Landrace × Yorkshine)) early-weaned piglets were used in this research to study the influence of PC on the growth performance, biochemical indexes, serum levels of IgA, IgG, IgM, IL-1, IL-2, IL-6 and gene expression of IGF-I in longissimus muscle, of IL-1 in jejunum mucous and mesentery lymphatic node, and intestinal microflora on animals. The objective of this study was to uncover the relationship of the function and structure of PC and the mechanisms of the influence of PC on the growth, immunity function and intestinal microflora of animals. This study contains three parts as follows:1.Effect of PC on growth performance and biochemical indexes of animals.A total of 36 Three-line crossbred (Duroc × (Landrace × Yorkshine))22day -early-weaned piglets were assigned to 3 treatment groups at randomly based on body weight, fed 3 diets respectively: 1. The control diets, 2. The low lever of PC diets (IP-control diet supplemented with 0.4% of PC, 3. The high lever of PC diets (HP- control diet supplemented with 0.8% PC). After two weeks feeding, the results shows as followed: In contrast to the control, there is a significant increment at ADG, at ADFI, at the activity of serum amylase and at the gene, expression level of IGF-I in longissimus muscle in HP (p<0.05); In contrast to the control there was no significant difference of the ADG and ADFI and the activity of serum amylase and gene expression level of IGF-I in longissimus muscle in LP (p>0.05); In contrast to the control there was a significant decrease at Blood Urea Nitrogen (BUN) of serum in high dose group(p<0.05); there was no significant decrease of serum level of BUN in low dose group(p>0.05). There was no significant difference in serum glucose, serum amino acid in three groups. All those indicate that high dose of polysaccharide from Cassiae seeds could improve the gene expression level of IGF-I in longissimus muscle of animals, increase the ability of digesting and absorbing starch, increase the synthesize of protein so as to improve the growth performance of animals.2. Effect of PC on serum antibody level, serum level of IL-1 P , IL-2, IL-6 andIL-1 3 gene expression in jejunum mucous and in mesentery lymphatic node of animals.To study the influence of PC on the immune system of animals, the same group design as experiment 1 was used. After two weeks, the results showed as followed: compared with the control, high dose group enhanced the content of IL-1 p\ IL-2 and IgG (P<0.05), especially IgA and IgM (PO.01) in serum, enhanced IL-1 £ gene expression in jejunum mucous (P<0.05) and especially in mesentery lymphatic node (PO.01); there was no significant difference in content of IL-1 p, IL-2 and IgA, IgG, IgM in serum and IL-1 £ gene expression in jejunum mucous and mesentery lymphatic node (P>0.05) between the control group and the low dose group, but low dose group enhanced the content of serum IL-6 (P<0.05) Those results suggested that polysaccharides from semen cassia could enhance some cell-mediated immune responses and the humoral immunity of piglets by modulating the activity of macrophages, T sell and B cell etc.3. Effect of PC on intestinal microflora of animals in vitro and in vitro was carried out. The influence of PC on ecoli and lactobacillus in vitro and on intestinal microflora of animals in vivo was analyzed by tranditional method and denaturing gradient gel electrophoresis (DGGE) analysis of 16S ribosomal RNA. The results showed as followed: In vitro, compared to the control, the number of lactobacillus was increased when the concentration of PC was higher than 0.05%, up to the highest at 0.8%; Compared to the control, there was no difference in the number of coliform in all different concentration of PC when only inoculated ecoliOg, but the number of lactobacillus was increased and the number of coliform was decreased at the same time, at which the concentration of PC was higher than 0.05% when inoculated the Fecal of piglet.In vivo, the intestinal microflora was examined from different groups piglets in animal trial as above, high level of PC could increase the number of lactobacillus and decreased the number of coliforms. It indicated that polysaccharides from Cassiae seeds could improve the prebiotics of animal intestinal microflora...
