Semen Cassiae is the dried ripe seed of Leguminosae plant Cassia obtusifolia L. or Cassia tor a L., which is being recorded in China pharmacopoeia. Semen Cassiae is sweet, bitter and salty taste, at the same time slight cool in nature. It converges to liver, kidney and large intestine channel. The efficacies include removing heat from the liver, improving eyesight, moisturizing intestines, relaxing bowel.Semen Cassiae has the exact function of reducing blood lipid. Recently, Semen Cassiae and its compounds preparations have been widely used to cure hyperlipidemia in clinical. At present, seldom have the pharmacological effect substance study, all can see are just plant and extracts. Have not seen the report about effective components of cure hyperlipidemia.The research adopt hyperlipidemia model by mice to study the pharmacological effect of the different extracts from Semen Cassiae. The result shows that the n-BuoH extracts have obvious action of reducing blood lipid. It can notably lower total cholesterin (TC) and glycerinum( TG ) in serum from mice of hyperlipidemia. So it can be determined that n-BuoH extracts is the active part.Take the methods of Macroporous adsorption resin, Silical gel column, Rp-C18 column and Sephadex LH-20 column chromatography to separate and purify the n-BuoH extracts. Six compounds were isolated and identified as obtusifolin, aurantio-obtusin, Gluco-obtusin, Gluco-obtusifolin, Daidzin and Cassitoroside by UV, IR, MS, 1H-NMR and 13C-NMR. Daidzin was isolated from Semen Cassiae for the first time.Develop the LDLR reporter gene system to research the action of reducing blood lipid of the six compounds. Result shows compound II could enhance the gene expression of LDLR and could reducing blood lipid.compound â…¡ is indicated to be the specific component in Semen Cassiae by the study of thin layer chromatography and high performance liquid chromatography.We get the specific component in Semen Cassiae by the research.
