Objectives To explore the expression of angiotensin II type 1 receptor(ATl) in human hepatocellular carcinoma (HCC) tissues and analyze its correlation with the pathological character of HCC tissues.Methods 27 fresh HCC tissues,27 adjacent liver tissues which are 2 centimeter distance to focus and 9 normal hepatocellular tissues are collected in the aflBliated Taihe hospital of Yunyang medical college from Jan,2003 to Jun,2004. Pathological stage and histological grade are carried out in HCC cases. Angiotensin II type 1 was measured in frozen sections of hepatocellular cancer tissues, adjacent liver tissuesand normal hepatocellular tissues by immunohistochemical staining and Western blotting. 10 visual field are observed in 10×40 high power lens in all sections.The scores are marked according to the strength and percentage of the staining cell, when score is over 10 the case is positive.then the score of hepatocellular tissues, adjacent liver tissues and normal tissues were analysed. Western blotting was employed to determine the expression of AT1 in 55 HCC tissues and their adjacent liver tissues,normal tissues.Results We found overexpression of AT1 in 23 cases(85.2%) among 27 hepatocellular cancer tissues,in 5 cases (18.5%)among 27 adjacent liver tissues, in 0 cases (0%) among 9 normal hepatocellular tissue. The positive staining is in cytomembrane and cytoplasm. The difference of positive rate of AT1 expression between hepatocellular cancer tissue and adjacent liver tissues, normal hepatocellular tissue was significant(P<0.01).The expressive strength of AT1 rises with clinic stage. Expression of AT1 in hepatocellular carcinoma was correlated with differentiation. When the degree of hepatocellular carcinoma is low,the score of AT1 is high.(P>0.05) . The expression of AT1 in the HCC tissue was significantly higher than that of the normal tissue and the adjacent liver tissue (P<0.05). OD value of the HCC tissue (2.01 ±0.12) is three times that of their adjacent liver tissue (0.25±0.15) .Conclusions The expressive of AT1 in hepatocellular cancimoma is high andcorrelated with pathological stage and histological grade. The high expression of ATI is an important maker of prognosis in hepatocellular cancinoma. ATI is a possible new target for treating hepatocellular cancinoma.
