Objective To investigate the effect of TGF-β1 on the proliferation and invasion of human colon carcinoma cell.Methods Two human colon carcinoma cell lines SW480 and LS174T were cultured in vitro. After exposed to different concentration of TGF-β1, the proliferation of SW480 and LS174T cells was measured by MTT. The expression of Smad4 and TGF-βR II protein of SW480 and LS174T cells were detected by immunocytochemistry. The migration and invasion were determined by BD BioCoat? Matrigel? Invasion Chambers.Results The proliferation of SW480 and LS174T cells treated 24 hours with TGF-β1 of different final concentrations (0.1μg·L~-1, 1.0μg·L~-1, 10.0μg·L~-1) have no significantly change compared with that of the controls (P>0.05). The invasion capacity of SW480 cells treated 24 hours with TGF-pl of different concentrations (0.1μg·L~1, 1.0μg·L~-1, 10.0μg·L~-1) have significantly increased compared with that of the controls (P<0.05). The invasive rates of SW480 cells treated 24 hours with TGF-pl of concentrations of 0.1μg·L~-1 1.0μg·L~-1 and 10.0μg·L~-1 were 30.2%, 41.1% and 41.7%, respectively. The invasive rates of SW480 cells treated without TGF-pl was 18.6% (P<0.01). The phenomenon was not observed in LS174T cells. SW480 expressed TGF-PRII protein but did not express Smad4 protein. LS174T cells expressed Smad4 protein but did not express TGF-PRII protein.Conclusion TGF-pl have no effect on proliferation of SW480 and LS174T,but TGF-pi can promoted the invasive capability of SW480 and this effect was independent with Smad4 signaling pathway. The effects of growth inhibition and pro-invasion of TGF-pi might be carried out via different signaling pathway.
