Objective: 1. To explore the prevention and cure of Naloxone on alcoholic fatty liver in rats and its mechanism; 2. To explore the effect of Naloxone on the content of 3 -endorphin in plasma and liver tissue in alcoholic fatty liver in rats; 3. To provide a efficiency method for the prevention and cure of alcoholic fatty liver in clinic.Methods: Forty-eight Wistar male rats were fed for one week normally, and then withdraw 12 rats for normal control group in random, intragastric infusion of normal saline by the dose of 15ml.kg~-1 once each morning, fed with common diet; Other 36 rats were administrated 15% liquor (diluted from 52% liquor) by the dose of 15 ml.kg~-1 from the first day to the three day, once each morning; and then were randomly divided into 3 group: model group(n=12) ,low dose Naloxone group(n=12), high dose Naloxone group(n=12). All of the rats in the last three group were administrated 52% liquor(intragastric infusion) by the dose of 15ml.kg~-1 from the fourth day, once each morning, feed with special diet(high fat and iron diet). All of the rats intake of water freely from beginning to end. The rats of four groups were treated the following measures after about one hour after intragastric infusion: the model group and normal group were treated with normal saline by the dose of 5ml.kg~-1 through abdominal cavity Injection respectively; the low dose Naloxone group and high dose Naloxone group were treated with Naloxone (0.016mg.kg~-1, 0.08mg.kg~-1, respectively) through abdominal cavity. And by the end of the fourth week, all rats were weighted, nartosised, sacrificed. The whole liver was gotten, rinsed repeatedly, sucked with filter paper, and weighted. And then measure SOD, MDA and β -EP of plasma, SOD and MDA of liver tissue. Liver sample were analyzed for histopathology and immunohistochemistry with microscope.Results: Rat models of AFL were established successfully. The level of plasma SOD increased while MDA and 3 -EP decreased in low doses treatment group and high doses treatment group markedly contrast with model group (P<0.01). The degree of hepatocyte fatty degeneration of low doses treatment group and of high dosestreatment group is more alleviative contrast with model group' and approaches to normal control group'. The amounts and distribution district of P -EP positive cell are consistent with the degree and distribution district of fatty degenerate hepatocyte.Conclusions: Long time drinking or excessive drinking can lead to AFL; Naloxone can prevent the formation of AFL by decreasing the level of P -endorphin (P -EP) in plasma and anti-lipid peroxide oxidation, and the result is dose-effect relationship; 3 -EP can become a new target for the prevention and cure of AFL.
