| Borneol named camphol in antient times, synthetized borneol contains borneol and isoborneol. There are a lot of pharmaceutical effect of borneol usedsingly. In the circulatory system the effect of borneol is similar to Guanxinshu Mixture Pill in dog with acute myocardical infaction;which can raise the blood volume in coronary venous fistula, reduce the heart rate and cardical oxygen requirement;in central nerve system borneol act as two -side action, which is inhibition or excitation. In addition, borneol can anti-inframmation, anti-bacterium, and cure the pain. Particularly, borneol always used as a assistant which has verified by modern medicine. Paeonol is the active component in Chinese drug paenia suffruticosa and cynanchum paniculatum, which was used widely in cardiivascular system, bacterium inframmation, cancer therapy, allergy disease and immune system. In central nerve system paeonol is not a addictive analgesics compared with morpine, which also can anti-pyrexia and protect the brain with insufficient blood. In this thesis the emphasis is on the effect of boeneol on the paenol pharmacokinetics and its distribution in mice brain and whether the dosage of borneol was associated with its action. The experiment was carried out in the fields as follows:1 .GC-MS method was established to detect the concentration of paeonol in mice plasma and brain. The paenol pharmacokinetics and its distribution in mice brain after i.g. had studied. The result of experiment was that paeonol was absorbed quickly in mice after i.g and eliminated by two-compartment model, and paeonol bioavailability was high. In addition, the concentration of paeonol in mice brain was low, which suggested it was difficult for paeonol to penetrate the blood-brain barrier.2. GC-MS method was established to detect the concentration of paeonol and borneol simultaneously in mice plasma and brain. The paenol pharmacokinetics and its distribution in mice brain effected by borneol had studied. The borneol pharmacokinetics and its distribution in mice. had also studied. The result of experiment was that borneol had little effect on the concentration of paeonol in plasma. On the contrary, borneol could promotepaeonol to distribute in mice brain, which suggested borneol could promote theopening of the blood-brain barrier.3. The paenol pharmacokinetics and its distribution in mice brain with different dose of borneol had studied. The result of experiment was that borneol in low dose or in ligh dose had little effect on the concentration of paeonol in blood compared with paeonol alone, but it didn't do the same as which in the brain. The mean concentration of paeonol at different time with low borneol in the brain was higher than which with high borneol, that is the dose of borneol isn't linear to the opening of blood -brain barrier, the reason hadn't certified. |
