Study on the relationships between NPY's expression and pituitary adenoma's proliferation and invasiveness and some kinds of hormone secreting of pituitary adenomasNeuropeptide Y, a 36-amino acid member of the pancreatic polypeptide family is a peptide originally isolated from porcine brain tissue by Tatemoto et al in the 1982,which turn into a form of barrette, so that can combine its receptor easily,"Y" refer to the tyrosines in the double end of it. It has the effect of hormone and neurotransmitter,widely distribute into the central and peripheral nervous system and other tissues and apparatus of circumambient,sach as the respiratory, circulatory, digestive and urinary system, it co-exist with noradrenalins in the sympathetic nerve,can contracting blood vessel intensely.NPY has widely physiologic functions,such as adjust hormone secreting of hypothalamus- pituitary-gland axes,occur and develop of pituitary adenoma, contract and diastole of smooth muscle, sober, emotion, blood pressure ,rhythm of the heart, breath and feed regulation, balance of energy, biothythm and feeding behavior,et al.It exerts important effect in hypertension, coronary heart disease and heart failure,hasing intimate relationship with study and memory, participating in the attack of dementia, depression and epilepsy.In this experiment,we collect 53 specimens of pituitary adenom which were embedded with paraffin,there are 25 cases of invasive pituitary adenomas and 28 cases of noninvasive pituitary adenomas in them,all of them are sliced into 4μm in series,after HE staining and immunohistochemical staining,then calculate the ratio of positive sample of NPY and calculate the ratio of cells which contain Ki-67 proteinin all of the cells through the observing with microscope, i.e. Labelling Index, LI, LI= cells which contain NPY protein and Ki-67 protein/ all of the cells X 100%.In the end,we discuss the relationship of NPY and Ki-67 with the invasiveness of pituitary adenoma,the relationship between proliferation and invasiveness,and the relationship among some kinds of hormone secreting of pituitary adenoma. Our experiment get the following results:1. The general positive ratio of NPY is 50.94%,it has 48% positive ratio in the invasive pituitary adenomas and 53.57% positive ratio in the noninvasive pituitary adenomas ,after statistical treatment,there are no significant difference of NPY positive ratio between invasive pituitary adenomas and noninvasive pituitary adenomas;The LI of Ki-67 in the invasive pituitary adenomas is (3.06 ± 1.55),the LI of Ki-67 in the noninvasive pituitary adenomas is (1.28 ± 0.86),after statistical treatment,there are significant difference of Ki-67 LI between invasive pituitary adenomas and noninvasive pituitary adenomas,the LI of Ki-67 in the invasive pituitary adenomas is higher than it in the noninvasive pituitary adenomas.2. There are no significant difference in the LI of Ki-67 between positive groups and negative groups of NPY.3. There is significant difference in the positive ratio of NPY among some kinds of hormone secreting of pituitary adenomas.LH is the highest, is 83.33%, next is FSH, is 75%,PRL is the lowest, is 11.11%.According as the result of experiment above,we conclude some cursory conclusion:1. There are osculatory relationships between the proliferation and the invasiveness,following the promoting of proliferation, the invasiveness is enhanced.2. NPY acting as a regulative peptide throughout the central and peripheral nervous system and other peripheral systems, it may not has a stimulative action inproliferation and invasiveness of pituitary adenoma, it may be a protective factor. 3. NP Y may has a selectively regulative action to some kinds of hormone secreting of pituitary adenoma.
