Human ether-a-go-go-related gene potassium channel (HERG1potassiumchannel), expressed in tumor tissues with various origination, have drawn muchattention in recent years for its correlation with the proliferation, apoptosis,invasion and some other characters of tumor cells. Our previous researchconfirmed the expression of HERG1in bone tumors with different malignancydegrades, and indicated the correlation between HERG1expression andpathology degrades. In this study, we investigated the relationship betweenHERG1expression and tumor invasion in Osteosarcoma, and explore its possiblemechanism.Objective: To investigate the expression of HERG1in SOSP-9607and MG-63cell lines, and uncover the role of HERG1in proliferation and invasion ofosteosarcoma cells by inhibiting the expression of HERG1using shRNA vectortransfection.Methods:(1) Study the expression of HERG1in SOSP-9607and MG-63celllines using immunocytochemistry, immunofluorescence staining and WesternBlot.(2) Construct the shRNA expression vector against HERG1.(3) Establish a stable cell line with reduced expression of HERG1using shRNA vectortransfection.(4) Study the proliferation of the Osteosarcoma cells with reducedHERG1expression using CCK-8assays.(5) Investigate the invasion ofOsteosarcoma cells with HERG1expression using transwell assays.Results:(1) HERG1is expressed in both SOSP-9607and MG-63cell lines.(2)The eukaryotic expression vector of HERG1shRNA was successfullyconstructed.(3) An Osteosarcoma cell line with reduced HERG1expression wasestablished using shRNA transfection technique.(4) CCK-8assay revealed thatdown-regulation of HERG1inhibited the proliferation of OSP-9607and MG-63cells.(5) Transwell assay revealed that down-regulation of HERG1inhibited theinvasion of OSP-9607and MG-63cells.Conclusions: HERG1is expressed in both SOSP-9607and MG-63cell lines.Down-regulation of HERG1expression in SOSP-9607and MG-63cells inhibitedthe proliferation and invasion of these Osteosarcoma cells. These resultsindicated that HERG1may play an important role in promoting the proliferationand invasion of Osteosarcoma cells.
