Objective To research the proliferative and invasive effect of TGFβon human adenocarcinoma cell line A549, and explore the underlying mechanism. Uncover tbe relationship of the degree of malignancy and TGFβ, providing experimental basis for the gene target therapy of lung cancer.Methods Inhibit or promote the TGFβeffect by transfection of siTGFβR2 and adding TGFβ1 in the cell culture respectively, examine the cell proliferative rate by MTT, examine the apoptosis degree by FCM, western blot test the expression of related protein, use wound healing to measure the cell migration ability, Transwell cell measure the cell invasive ability.Results 1. TGFβ1 promotes the proliferation of A549, and inhibits its apoptosis; siTGFβR1 may inhibits cell proliferation. 2. TGFβup-regulate the expression of PI3K. 3. TGFβpromotes the invasion and motility of A549, inhibits the expression of E-Cadherin, whereas increase the content of MMP-2,MMP-9. 4. TGFβincreases the phosphorylated Erk1/2, Erk1/2 inhibitor can reverse the effect mentioned in item 3. Conclusion TGFβcan accelerate the proliferation and invasion of A549, the promoted proliferation may connect with activated PI3K/AKT signalling, the enhanced invasion may due to the over expression of MMPs and inhibition of E-Cadherin mediated by Erk1/2.
