IntroductionThe telomerase inhibitors maybe be a kind of anti - cancer medicine that can kill tumors cells. The research on the inhibition effect of telomerase siRNA on the growth of human oral cavity squamous cell carcinoma provides vast applied foreground for the development of new drugs and the prevention of diseases. The correlationship between c - fos and hTERT is still unknown. The c -fos and c - jun constructs the dimmer forming AP - 1 ( activator protein - 1 ) complexes which combine with the DNA to affect the transcription of many factors , regulates the expression of late reactive genes and the transcription activity, controls the cell growth cycle. Ameloblastoma ( AB) is the most frequently occurring odontogenic tumor,and it may progressively invade the jaw, recur locally and transform malignantly. This study detected the expression of c -fos, c - jun mRNA and their relationship with hTERT in AB. And the study analyzed the relationship between the expression of c - fos, c - jun mRNA and the clinical pathology of AB.Materials and methodsThe specimens be used by in situ hybridization were surgically removed from 54 patients with AB (primary AB 31 cases, recurrent AB 17 cases, malignant AB 4 cases) and 7 patients with normal oral mucosa at the First Affiliated Hospital of China Medical University between 1996 -2000. The technique of in situ hybridization was provided by Beijing University. We detected c - fos, c -jun, hTERT mRNA by in situ hybridization. The extent of staining was scored;( - ) fewer than 10% positive or no staining;( + ) 10% -40% positive;( ++ ) 40% -70% positive;( + + + ) more than 70% of cell positive. Adopted SPSS 11.5 statistics software to carry out x2 test and Fisher s accurate probability method and Kendall correlation analysis. P <0. 05 meant that the tissue specimens had statistical significance.ResultIn 7 cases of normal oral mucosa, 1 case of hTERT presented weak positive in epithelial basement membrane. hTERT of 51 cases in 54 AB cases were all positive, the expression ratio was 94.4%. The positive parts most located in the peripheral column or cubic cells and scattered or dense proliferating polygonal, spindle cells. The expression in AB and normal epithelium groups indicated significant variance ( P < 0. 001 ). The positive ratio and expression intensity of hTERT mRNA increased with the increase of pathological grading of recurrence and canceration of AB.Almost no expression of c - fos mRNA presented in 7 cases of normal oral mucosa. The positive expression in AB increased obviously to 92. 0% (46/50) , expression located in the periphery and reticular layer. Some cases presented nuclear positive, but granular cells and keratinoid cells were almost negative. Odontogenic epithelial rest was positive, and interstitial fibers, vascular endo-thelial cells and lymphocytes, plasma cells were positive. And the lacunar cells of bone tissue around the tumor and osteoclasts were positive. AB was compared with the normal oral mucosa, P <0.05.No expression of the c - Jun mRNA appeared in 7 cases of normal oral mucosa. The positive expression of c -Jun mRNA in AB was 27. 1% ( 13/48) , in which 4 cases were weak positive, and moderate and strong positive were 7 cases and 2 cases. Between normal oral mucosa and AB, P <0. 001;among the three groups, primary, recurrence and malignant AB, P>0. 05, indicated no statistical significance.The Kendall correlation analysis between hTERT mRNA and c - Fos mRNA, rk =0. 196, P =0. 035, presented high positive correlation. Between hTERT mRNA and c - Jun, rk = -0.648, P<0.001, presented moderate neg-ative correlation. Between c -Fos mRNA and c - Jun mRNA,rk = -0. 306, P <0.001, presented moderate negative correlation.Conclusion1. c - fos protein possibly constituted a cardinal subunit of AP - 1 to participate the regulation of the telomerase activity, so it was important in the occurrence, development and proliferation of AB.
