IntroductionMiddle—ear cholesteatoma is a proliferative form of keratinocyte dysregulation with aggressive growth that often leads to destruction of adjacent bone in the milddle ear cavity. However, the molecular mechanism underlying these pathological changes is largely unknown. Keratinocyte growth factor (KGF) belongs to a family of fibroblast growth factor (FGF). Previous study showed that the target cells of KGF is the endo-thelial cells, even though KGF is exclusively expressed in the mesenchy-mal cells. Keratinocyte growth factor (KGF) is a mesenchymal cell — derived paracrine growth factor that specifically stimulates epithelial cell proliferation, and KGF exerts its biological function by binding to its receptor, KGFR, which is expressed in the endothelial cells. Our study is to understand the possible involvement and relationship between KGF and KGFR in the pathogenesis and development of the middle — ear cholesteatoma. So far, there is no such a report in our country.Materials and Methods1. Materials: we collected 39 middle—ear cholesteatoma specimens as case group from cholesteatomaectomy which were operated between January and June in 2005 in the first affiliated hospital, China medical u-niversity. These specimens had been diagnosed as cholesteatoma by the routine pathological examination. We collected 10 epithelial cells samplesfrom the external auditory meatus as control group. These samples were either from the discarded external auditory meatus skin from the ear deformities plastic surgery or from the skin around the external auditory canal tumor by tumor ectomy. These samples were diagnosed as normal skin tissue by pathological examination.2. Methods-, we examined the expression of KGF and KGFR protein by SABC immunohistochemical staining.3.
