Experimental Study On The Effect And Mechanism Of Compound Biejia Ruangan Prescription On Glomerular Sclerosis

The renal fibrosis is often the common pathological character of most renal diseases. It a complicated process and involves the production cytokine, adhesion molecules, chemotactic factors,GMCs and fibroblast's hyperplasy and the accumulation of ECM,ect.The ECM in kidney is composed of fibronectin, laminin and collage. ECM has important effect on the physiological function.It not only promote the adhere of cells,but also infect their metabolism proliferation and differentiation.So it is important to research the ECM to cure the renal fibrosis.The CBRP is composed of carapax amydae,angelica, angelicaangelica, Codonopsis pilosula ect.It can softening hard mass, deintoxication, advantage gas, keep blood. It can inhibit the synthesis of collagen. According to the traditional Chinese medicine theory ,we use CBRP to cure the renal fibrosisObjectiveTo study the major pathological of experimentcal renal fibrosis.We use the 5/6 nephrectomy rats,and expect to find the theoretical base from the in vivo and ex vivo experiment.Methods90 male SD rats, weighting 200±20g, adopted for a week. To investigate glomerular sclerosis occurs and therapeutic effect of CBRP on GS,the Animal model was established by 5/6 nephrectomy and the rats were divided into 8 groups:normal group has 10 rats. sham operation group has 8 rats. model group,Benazepril group,Niaoduqing group, CBRP low , middle and high dose groups, and each group has 12 rats.Benazepril and Niaoduqing are positive medicine control groups.From the day after modeling, CBRP solution of different dosages(1.4g/kg,0.7g/kg,0.35g/kg)was given to rats in the high, moderate and low CBP group by gavage respectively, at the same time equal volume of normal saline was given to those in the normal control group, sham-model control group and model control group, and an equal volume of prednisone was given to those in positive medicine control group.
