Objective To study the expressions of Vascular endothelial growth factor(VEGF)and cyclooxygenase-2 in transitional cell carcinoma of the bladder (TCCB) and analyze their correlation with the biological behavior of the tumor.Methods The expressions of VEGF and cyclooxygenase-2 in 68 cases of TCCBand 10 cases of normal bladder tissue were detected by streptavidin-peroxidase immunohistochemical method.Results VEGF was negatively expressed in normal bladder mucosa.The positiveexpression rates of VEGF were 23.07% ,58.62% and 84.61% for G1, G2 and G3, 43.75% and 77.77% for TisT1 and T2T4 tumors ,respectively. The positive expression rates in three groups with different histological grading in TCCB showed significant difference(X2=14.1, P<0.01),and the difference was great significant as well in two different clinical staging(X2=8.29, P<0.01);The expression of VEGF in recurrence group and unrecurrence group was 78.94% and 40.00%,respectively,and the difference was significant (X2 =10.77, P<0.01). COX-2 was negatively expressed in normal bladder mucosa.The positive expression rates of COX-2 were 15.38% ,55.17% and 80.77% for G1, G2 and G3, 37.50% and 75.00% for TisT1 and T2T4 tumors .respectively. The positive expression rates in three groups with different histological grading in TCCB were significantly different (X2=15.27,P<0.01),and the significant difference was found as well in two different clinical staging (X2=9.73, P<0.01);The positive expression of COX-2 in recurrence group and unrecurrence group was 73.68% and 36.67%, respectively, and the difference was significant(X2=9.40, P<0.01).The expression of VEGF in TCCB had a close correlation with that of COX-2(r=0.55, P<0.01).Conclision The positive expression of VEGF and COX-2 are closely related to thetumor-cell grade, depth of invasion and recurrence .Both the expressions of VEGF and COX-2 may be used as parameters for assessing the malignancy and recurrence of TCCB.
