Qingyangshen also named Cynanchum otophyllum is one of the traditional Chinese medicine which is widely used in some diseases such as chronic hepatitis, tuberculosis and epilepsy et al. Qingyangshen is a plant of the genus Cynanchum L (Asclepiadaceae) and it is distributed extensively over Southwest China. The coworkers from Chinese Academy of Sciences have extracted some effective component using Chloroform. In the study we are focusing on the effects of the chloroform extracts to the chronic amygdala kindling model. Observing the Racine's stage and the EEG is also required in the experimens. Also recording the EEG changes and ADD is needed to get the results about the effects on the Kindling model.Objective:To study the effects of Qingyangshen extracts - Qingyangshenylycosides on the ideal chronic epilepsy model(amygdala electrical stimulated kindling model), using the solvent as the control to compare the effects.Methods: Amygdala Kindling models are established in rats to observe the influence of Qingyangshenlycosides on the kindling development and kindled rats. The Afterdischarge duration(ADD) and the EEG changes are also recorded. Also the influences on the rats Racine's stage are also observed. This present study is designed to investigate the effects and possible antiepileptic mechanism of Qingyangshenylycosides on amygdala electrical kindling in rats. Results:1. Amygdala kindling was established successfully. Qingyangshenlycosides 6.25mg.kg-1 intraperitoneal can significantly retard the presence of Racine's Stage V seizure and the development of amygdala kindling (P<0.01).2. Qingyangshenlycosides 12.5mg.kg-1 and 25mg.kg-1 can reduce the Racine's stage of kindled rats,shorten the afterdischarge duration(ADD), inhibit amygdala kindled seizure in rats.3. Qingyangshenlycosides 6.25mg.kg-1 ip combining with valproate sodium 200mg.kg-1 ig can reduce the afterdischarge duration(ADD), lower the Racine's stage (P<0.05).4. Qingyangshenlycosides 6.25mg.kg-1 ip combining with nicardipine 2 mg.kg-1 ip can only shorten the afterdischarge duration(ADD)(P<0.05) and have no significant influence on the Racine's stage.5. Qingyangshenlycosides 50mg.kg-1 have no effect on the maximal electroshock (MES) in mice when using individually, but it can significantly prolong the latency of convultant mice, shorten the duration time of the convultant mice when using combined with valpronate sodium200mg.kg"1(P<0.05).Conclusion: Qingyangshenlycosides(6.25mg.kg1 ip) can inhibit the amygdala electrical kindling development in rats and prolong the developing time of kindling obviously. Meanwhile Qingyangshenlycosides(12.5mg.kg"1, 25mg.kg"' ip) inhibit the electrical kindled rats, reduce the Racine's stage, shorten the ADD. Qingyangshenlycosides combined with other chmical drugs can inhibit the seizures in kindled rats. Also combination of Qingyangshenlycosides and valpronate sodium have effect on the maximal electroshock(MES) mice model. All these results suggest that Qingyangshen chloroform extracts may have the important significance to prevent and treat epilepsy as one traditional Chinese medecine.Graduate student:Wu Pei (Pharmacology) Directed by Yue Wang...
