Experimental Study Of Oxymatrine In Preventing Renal Glomerulosclerosis In Adriamycin-induced Nephropathy Rat

Objective:To study the mechanism of the effects of oxymatrine on chronic glomerulosclosis in the kidney of adriamycin-induced nephrcpathy rats .Method:150 male Wistar rats of were randomly divided into five groups:A group (controlgroup); Bgroup(modelgroup); C group(benazepril treatedgroup);Dgroup(OM 50mg/(kg. d)) ;Egroup((OM 100mg/(kg.d)). Rats in A, B, C and D groups were given adriamycin 2mg/kg intravenously on the 1^st and 21^st day respectively. After 7 weeks , five rats in each group were killed every 4 weeks for blood biolchemical analysis and histological study. The expressions of Matrix metalloproteinase -1 (MMP-9) Tissue inhibitor of metalloproteinase-1 (TIMP-1) transforming growth factor—β1 (TGF—β1) connective tissue growth factor (CTGF) were measured with immunohistochemistry method.Results:(1)24h Upro excretion in B group was higher than those in other groups at each time point during the experiment (p<0.01) , After 15 weeks, Cr and BUN of B group were all higher than those in other groups(p<0. 01); (2)By 23 weeks of the experiment,the glomerular sclerosis scores in C D, E groups increased remarkably comparing with the A group (p<0.01) ;(3)By 11 weeks, immunohistochemistry staining indicated that there was an significant increasing TGF—β1,CTGF, expression in B group as compared to the three treatment groups. After 19 weeks,the expressions of MMP-9 in B group was lower than those in A,B,C and E groups (p＜0.01),and TIMP-1 in B group was higher than those in other groups (p＜0.01).Conclusion: OM can inhibt the progress of glomerular sclerosis in the kidney of adriamycin-ibduced nephropathy rats.
