Glioma is one kind of common maglinant tumors in centralnervous system, and it threatens the health and lives of peopleseriously, about 40% of the brain tumors are glioma. Glioma has beenconsidered as one of the most difficult maglinant tumors to be cured.At present, the average suivival time for maglinant glioma patients isstill less than one year. Nowdays the effective treatment for gliomapatients includes operation,chemotherapy,radiotherapy,genetictherapy etc. With the appearment of new chemotherapy drug,chemotherapy is becoming more and more important in the field ofglioma therapy.Elemene, the new second-class antitumor drug which is isolatedfrom the Chinese herb Rhizoma Zedoariae, was shown to exhibitantitumar activity. In 90s, the elemene was applied to treatment ofbrain tumors, and an aspiring result was got. It can pass across theblood-brain-barrier, may improve the cellular immune function, hasno drug resistance. With the advantage of low toxity to heart,liver etc,high efficacy and broad-spectrum, the elemene is paid more attentionsby medical field, and it is broadly used to the treatment of multipletumors, including maglinant glioma. Now, people pay more attentionto the usage of elemene in the clinical therapy, there is little studyabout the anticancer mechanism of elemene.Objective To investigate the proliferation inhibitory effect ofElemene on rat C6 glioma cells and the inhibitory mechanismbetween them. To observe the effect of immunity regulation ofElemen.Methods The proliferation inhibitory effects of Elemene on ratC6 glioma cells and spleen thymus-depenent lymphocyte were studiedby using MTT assay, and then analyzed apoptosis and cell cycle of ratC6 glioma cells by flow cytometry (FCM). The models of C6tumor-bearing mice were produced and treated with Elemene, Thelevel of IL-2 and the NK cell activity were measured by using MTTarrays.Result The proliferation of C6 glioma cells were significantlyinhibited by Elemene, and with the increase of drug concentration andthe elapse of time, the proliferation inhibitory effect increased too.When Elemene concentration was between 10ug/ml and 30ug/ml,Elemene can stimulate the proliferation of spleen thymus-dependentlymphocyte;when the concentration was more than 40ug/ml, theproliferation of spleen thymus-dependent lymphocyte wassignificantly inhibited. FCM revealed that apoptosis cells increasedwith the increase of drug concentration and the elapse of time. Cellcycle transition arrested from S to G2/M phase was noticed. The levelof IL-2 and NK cell activity were significantly lower in C6tumor-bearing mice than control group;after treated, the level of IL-2and NK cell activity increased significantly.Conclusion Elemene can significantly inhibit the proliferationof C6 glioma cells by inducing the cell apoptosis, and Elemene causeda dose-dependent and time-dependent proliferation inhibitory effect.Elemene could obviously promote the level of IL-2 and NK cellactivity, could significantly enhance the immunity function oftumor-bearing mice.
