Coronary heart is the most principal disease that affectshuman death in China, and its pathogenesis is complex. Recentstudies have showed that oxidative stress is one of the importantreasons to cause coronary heart.Hawthorn,a kind of plant belonging to rosaceae crataegus,is a traditional herb in China. Now, hawthorn is widely used tocure coronary heart disease, especially flavonoids constituent,thebasis of hawthorn, which play a significant pharmacologicalrole in the therapy of coronary heart disease.To observe protective effects of hawthorn leaves flavonoidsin oxidative stress injury, we Cultured cardiocyte of neonatal rats,and stimulated them by hydrogen peroxide (H2O2),and thenexamined cardiocyte morphology,death rate,biochemistry indexand apoptosis of cardiac myocytes by microscope,typpan bluestaining , biochemistry analyses , DNA agarose gelelectrophoresis.and flow cytometrymethods.We found numerous fragmentation and crimple ofcardiocyte after incubation with 0.5 mmol/L H2O2, at the sametime, improved LDH,CK release, increased MDAgeneration,decreased SOD and mitochondria dehydrogenaseactivity ,however rapid growth and clear configuration incontrast cells. The present study demonstrated that the cardiocyte deathis significantly associated with lipid peroxidation , inhibitingSOD activity and mitochondria damage.We farther study protective effects of hawthorn leavesflavonoids on oxidative stress injury. We found that hawthornleaves flavonoids decreased cadiocyte death rate induced byH2O2 and reduced LDH,CK release. simultaneity ,we foundhawthorn leaves flavonoids can effective decrease MDAgeneration,increase SOD activity and improve mitochondria.dehydrogenase activity. The result demonstrated that protectiveeffects of hawthorn leaves flavonoids is significantly associatedwith decreasing Membrane lipid peroxidatic reaction andincreasing antioxidase activity.Furthermore, the study showed oxidative stress andnumerous free radical caused cadiocyte apoptosis. Our studydemonstrated hawthorn leaves flavonoids can effectively inhibitcadiocyte apoptosis induced by H2O2 , but the mechanism willstudy in future.The present study demonstrated that hawthorn leavesflavonoids can prevent cardiocyte injury from oxidative stress,and showed the mechanism. The study provide the evidence intheory on preventing and cureing coronary heart disease inducedby oxidative stress.
